Objectives: Grape procyanidins (GPC) are natural polyphenols extracted from grapes. It was reported that GPC have a lot of bioactivities such as antioxidation, antiradical, antitumor, and cardioprotective effect. In this study we investigated whether GPC can protect oxidative injury of human umbilical vein endothelial cell strain (ECV304) induced by free fadicals in vitro.Methods: 1. In vitro it takes a model of endothelial cell oxidative damage induced by Fenton reaction produced oxygen-derived free radicals, Adriamycin (ADR) and ultraviolate, respectively. MTT assay was used to test influence of GPC on free radicals induced inhibition of cell proliferation.2. In vitro oxidative damage to ECV304 was caused by Fenton reaction, reverse transcription-polymerase chain reaction (RT-PCR) was used to analysis influence of GPC on OH induced abnormal expression of endothelial cell ICAM-1 mRNA.3. Flow cytometry was used to analysis endothelial cell apoptosis induced by Fenton reaction produced hydroxyl free radical.4. Malanyldiadehyde (MDA) and superoxide dismutase (SOD) was analysised by chemical methods, on which GPC influenced when ECV304 was damaged by OH.Results: 1. In positive groups damaged by Fenton reaction, ADR and ultraviolate, repectively, Cell proliferations decreased in different degrees. While after pretreated by GPC, cell proliferation damages lowered obviously, cell proliferations in some groups even higher than normal controls.2.Compare with normal control, ICAM-1 mRNA expressions in groups pretreated with GPC decreased remarkably, and decrease with GPC's increase. mRNA expression of GPC control was the lowest. Areas and brightnesses of electrophoresis belts and inhibition rates caculated according to these were coincide with electrophoretogram.3. The result of flow cytometry shows apoptosis rate of positive control is higher than normal control, cell apoptosis rates decreased remarkably after pretreated by GPC, and they decrease with increase of GPC dose.4.MDA testing result show that MDA levels of normal control, positive control, GPC control and GPC group(100ug/ml) are 0.979 + 0.0420, 0.699 + 0.0420, 2.361 +0.0818, 1.748 + 0.0531, respectively. In these, MDA level of GPC control is the lowest, the content of GPC group decreased than positive control remarkably. Conclusion: l.GPC can lessen peoliferation inhibition of endothelial cell induced by oxygen-derived free radicals, inhibit oxidation caused abnormal expression of cell ICAM-1 mRNA, decrease apoptosis and Lipid peroxidation products in cell. It was suggested that GPC have good protective effect againstin free radicals inducedendothelial cell damages.2.According to our studies on free radical scavenging effects of GPC in vitro, it is analysised that the mechanisms of anti-oxidation and proliferation promoting effects of GPC are closely related to their strong free radical scavenging and train reactions inhibiting capacity. In addition, by inhibiting expression of intercellular adherance molecular, GPC can decrease the adherance between endothelial cell and leukocyte, lessen local inflamation, so that protect arterial wall from being injuried. inhibiting formation of Atherosclerosis and thrombosis.
