Objective:It takes bone merrow cells as model, to investigate whether GPC can protect injury of mice bone merrow cells induced by oxidative injury and radiation injury in vitro and vivo.Methods: 1. The mice bone marrow cells given different dose of GPC were injuried by H2O2 , the proliferation of mice bone marrow cells was measured by MTT assay,the expression of Bax was measured by immunohistochemistry method.. The mice bone marrow cells given different dose of GPC were injuried by ~60Co γ -rays, the proliferation of mice bone marrow cells was measured by MTT assay.2. The mice given different dose of GPC were exposed to ~60Co γ -rays whole body subacutely. The proliferation of mice bone marrow cells was measured by MTT assay, the cell cycle and apoptosic rate were measured by flow cytometry method, the Bcl-2 and Bax expression were measured by immunohistochemistry method.Results: 1.In vitro of 6h and 24h, the cell proliferation of the 3 doseof GPC groups was much higher than those of the normal control groups(p<0.05) .The cell proliferation of the H2O2 injuried control groupswas much lower than those of the normal control groups, it was higher inthe GPC groups than those of the H2O2 injuried control groups(p<0.05 ).The expression of Bax was much higher in the injuried controlgroup than that of the normal control groups, it was much lower in thehigh-dose and middle-dose GPC groups than that of the injuried controlgroups(p<0.05) The cell proliferation of the irradiating control groups... |
