| Objective:To investigate distributional characteristics of two Gene Polymorphisms of Platelet Glycoprotein, GPâ…¢a PLA and GPâ… a807C/T in Han population of Qingdao, China. To study whether two Gene Polymorphisms contribute to aspirin resistance. And to analysis the probable correlated factors of AR.Methods:152 patients with artherosclerosis on aspirin (100mg/d) for at least 14 days were enrolled. According to the Platelet agglutination test, they were divided to three groups:aspirin resistant (AR) group, aspirin semiresponder(ASR) group and aspirin sensitive (AS) group. Recording gender, age, blood routine, blood fat, glucose, history of smoking, hypotension and diabetes of patients. Genotyping of the two polymorphisms was performed using a polymerase chain reaction-based restriction fragment-length polymorphism analysis.Results:8 patients (5.26%) showed AR. The rate of diabetes was higher in AR,ASR patients, compared with Aspirin sensitive patients (P<0.005). Only one patient carried the PLA1/PLA2 allele. The others possessed PLA1/PLA1 allele. None PLA2/PLA2 was observed. No relation was found between resistance status and PLA genotype. For the 807C/T genotype, the frequency of T allele in AR group and ASR group were higher than that in AS group. The difference was statistically Significant (P<0.005).Conclusion:The incidence rate of AR is 5.26%. Diabetes are the correlated factors of AR. The present results suggest that the PLA polymorphism of GPâ…¢a is not a risk factor for AR. The 807T allele of the GP I a gene show higher relationship with aspirin resistance. It may be a genetic factor for susceptibility to AR. |
