Background:Cisplatin is one of the most common drugs in the clinical treatment of cancer. Because of its strong and wide curative effect, it was recommended as the first choice for lung cancer, bladder cancer, testicular cancer, ovarian tumors and so on. However, while cisplatin exerted its effects on killing the tumor cells, it also produced some side effects in the body. This obstructs its clinical application in a great measure. Curcumin has been proved to have pluralism biological action and nontoxic, including anti-tumor effects and protect effects on liver and kidney. Combining curcumin with cisplatin will be a great potential therapy for tumors.Objective:To investigate the effects of curcumin on the injury of gastrointestinal impetus and structure caused by cisplatin in mice.Methods:40 male mice were randomly divided into four groups:normal control, cisplatin. cisplatin combined with curcumin 1 day and 15 days. The mice were orally administered with 0.2ml solvent solution (5% Arabic gel) once a day for 15 times in normal control, cisplatin and cisplatin combined with curcumin 1 day. The mice in cisplatin combined with curcumin 15 days were orally administered with 0.2ml curcumin (200mg·kg-1) in solvent solution (5% Arabic gel) once a day for 15 times. On the 12th day, the mice in cisplatin, cisplatin combined with curcumin 1 day and 15 days were given cisplatin by intraperitoneal injection (2mg·kg-1,0.2 ml, once a day for 4 times). At the same time, the mice in normal control were given saline by intraperitoneal injection in the same way. On the 15th day, the mice in cisplatin combined with curcumin 1 day were orally administered with curcumin in solvent solution(200mg·kg-1,0.2 ml).24 hours after the last injection, mice were orally administered with Indian ink in solvent solution(0.8ml). After 30 minutes, the gastric residual rate and the motional rate of small intestine were detected. The morphological changes of intestines were observed by HE dying.Results:In cisplatin group, the weight of mice was obviously decreased, the gastric residual rate was significantly increased, and the motional rate of small intestine was obviously decreased. In 1 day combination group, curcumin has no obvious effects on the changes induced by cispliatin, but in 15 days combination group, the weight of mice was raised, the gastric residual rate was obviously decreased, and the motional rate of small intestine was significantly increased. HE dying results:In cisplatin-treated mice, small intestinal mucosa of mice was necrosis seriously. Intestinal villus was not well-arranged. Partial cells in epithelial layer were necrosis (karyolysis). In the lamina propria, inflammatory cells infiltrated obviously. Partial glands of intestinal lamina propria were necrosis, and partial glands were atrophy. In cisplatin combination with curcumin 15 days'group, the necrosis of epithelial cells in intestinal mucosa was not obvious and only partial cell degeneration was seen. No significant necrosis or atrophy was seen in the glands of lamina propria.Conclusion:The pre-administration of curcumin can attenuate the gastrointestinal injury caused by cisplatin.
