Objective: To investigate the mechanism of curcumin gavage for relieving the dysfunction of gastric emptying induced by cisplatin whether through influencing the content of acetylcholine. To provide the necessary theoretical basis for future clinical application of curcumin, especially in reducing the side effects of cisplatin in gastrointestinal tract, also to provide theoretical basis for the development and application of curcumin in other gastrointestinal diseases.Methods: Thirty-two Kunming male mice were random Ly divided into 4 groups: Control, cisplatin, cisplatin+curcumin and curcumin. There are 8 mice in each group. Curcumin(200mg/kg)was treated once a day by gavage, a total of 15 days. The mice in other groups were orally administered with solvent solution in a same way as a control. After 24 hours for the last gavage, the gastric residual rates of mice in each group were detected. Then the mice were dislocated to death, and the stomach tissues of mice were cut out. The content of acetylcholine(ACh) and the activity of acetylcholinesterase(Ach E) in stomach were detected. The m RNA and protein expression of Ach E were further detected by real time relative quantitative PCR and immunohistochemistry individually. SPSS11.0 statistical software was used for variance homogeneity test, Q test(Newman-Keuls method) analysis and comparison among the different groups, P<0.05 as a statistically significant difference.Results: Compared with control group, the gastric residual rate of mice was increased significantly in cisplatin group(P<0.05). The content of Ach and the activity of Ach E of stomach decreased significantly in cisplatin group(P<0.05), and the expression of Ach E m RNA and protein was also decreased in cisplatin group(P<0.05). Curcumin gavage significantly relieving the increased gastric residual rate of mice induced by cisplatin(P<0.05). The changes of Ach and Ach E in stomach induced by cisplatin were decreased significantly by curcumin gavage(P<0.05), and the changes of Ach E m RNA or protein in stomach were also significantly decreased by curcumin(P<0.05). There is no significant changes in single curcumin treated group.Conclusion: Curcumin gavage can relieving the dysfunction of gastric emptying induced by cisplatin, and one of the mechanism may be through relieving the change of acetylcholine in stomach. There is no significant affects of curcumin on normal gastric emptying.
