Objective To explore the effects of visfatin in glucose and energy metabolism and hepatic insulin insistence by detecting the expression of visfatin in liver of rats in different glucose metabolic statuses.Methods SD rats were randomly divided into five groups:normal control group(group A,n=10),obesity group (group B,n=12),diabetes mellitus group(group C,n=10),diabetes controlled by insulin group(group D,n=9)and diabetes controlled by metformin group(group E,n=9). Rats in group A were fed with normal diet and other groups were fed with high glucose and fat diet. After 8 weeks, rats of C,D,E groups were injected streptozotocin (STZ,40mg/Kg) intraperitoneally to induce a model of diabetes mellitus (DM). The DM animal models were considered as success when rats'fasting blood glucose (FBG) were more than 16.7mmol/L twice. FBG of rats in group D were controlled by Isophane Insulin hypodermic injection and group E were controlled by Metformin Hydrochloride Tablets(500mg/kg.d) via gastric fistula until FBG lower than 11.1 mmol/L.12 weeks later, fasting body weight of rats in group B should be 20% heavier than the average body weight of group A. Then the rats were sacrificed, collected the data of blood glucose, measured serum lipid,FFA by biochemically and insulin by radio immunoassay. The liver of rats was used to measure visfatin,G-6-pase,PEPCK mRNA by RT-PCR and visfatin, AMPKα, p-AMPKαprotein by western blot.Results FBG of group C was significantly higher than group A and B (p<0.01);FBG of group D and E were significantly lower than group C (p<0.01).Fins and HOMA-IR of group B and C were significantly higher than group A (p<0.01);Fins and HOMA-IR of group C were significantly higher than group B (p<0.01).ISI of group B and C were significantly lower than group A (p<0.01);ISI of group C was significantly lower than group B(p<0.01).The level of serum TG of group B was significantly higher than group A (p<0.01);TG,TC,HDLC,LDLC of group C were significantly higher than group A (p<0.05); TG,TC,HDLC of group D and E were significantly higher than group A (p<0.05); TG,HDLC,LDLC of group C were significantly higher than group B(p<0.05); TG,HDLC,LDLC of group D and E were significantly lower than group C (p<0.05). The level of serum FFA of group C,D and E were significantly higher than group A (p<0.05);FFA of group C was significantly higher than group B(p<0.05).The expression of visfatin mRNA of group C was significantly higher than group A and B (p<0.05);visfatin mRNA of group D and E were significantly lower than group C (p<0.01).The expression of G-6-Pase mRNA of group B was significantly lower than group A (p<0.01);G-6-Pase mRNA of group D was significantly higher than group A (p<0.05);G-6-Pase mRNA of group C,D and E were significantly higher than group B (p<0.05);G-6-Pase mRNA of group E was significantly lower than group C and D (p<0.05).The expression of PEPCK mRNA of group B and D were significantly lower than group A (p<0.01);PEPCK mRNA of group C and E were significantly higher than group B (p<0.01);PEPCK mRNA of group D was significantly lower than group C (p<0.01);PEPCK mRNA of group E was significantly higher than group D(p<0.01).The expression of visfatin protein of group C,D and E were significantly higher than group A (p<0.05).The expression of AMPKa protein of group C,D and E were significantly lower than group A (p<0.05);AMPKa of group C was significantly lower than group B (p<0.05).The expression of p-AMPKa protein of group B,C,D and E were significantly lower than group A (p<0.01)Conclusion The expression of visfatin in liver of SD rats could influence glucose and energy metabolism, might associate with insulin resistance and diabetes.We could not consider that visfatin can influence the pathway of metformin activated AMPK to decrease blood glucose.
