Objective: To investigate the relationship between the allele frequencies and genotype distribution of single nucleotide polymorphism (SNP) at position -11377 in proximal promoter region of adiponectin gene and type 2 diabetic nephropathy in Han Nationality in Guibei , which are likely to provide a new means for the early diagnosis and gene therapy of diabetic nephropathy.Methods: Adiponectin gene -11377C/G polymorphism was examined using polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) in 42 healthy controls, and 108 patients of T2DM which contains of 50 type 2 diabetic patients with DN. Body height, weight, total cholesterol(TC), triglyceride(TG), high density lipoproteins(HDL), fasting plasma glucose(FPG), were measured. The statistical analyses were performed using the SPSS for Windows software (version 17.0).Results: The frequencies of CC,CG and GG genotypes of ANP SNP-11377 in healthy controls were 71.4%, 23.8% and 4.8% , respectively. The allelic frequencies for C was 83.3% and G was 16.7%. The frequencies of CC,CG and GG genotypes of ANP SNP-11377 in T2DM were 46.6%, 46.6% and 6.8% , respectively. The allelic frequencies for C was 69.8% and G was 30.2% . The frequencies of CC,CG and GG genotypes of ANP SNP-11377 in DN were 58%, 36% and 6% , respectively. The allelic frequencies for C was 76% and G was 24%. The CG+GG genetypes and G allele frequency distributions of SNP-11377 were higher in T2DM patients without DN than that in healthy controls (P=0.013,P=0.028). The distribution of genetypes and allele frequencies of SNP-11377 polymorphism had no statistical difference between T2DM patients with DN and T2DM patients without DN(P>0.05).The body mass index (BMI) was higher in CG+GG genetype than that in CC genetype in T2DM patients without DN. (P=0.037)Conclusions: SNP-11377C/G polymorphism of adiponectin gene is associated with type 2 diabetes in Han nationality in northern Guangxi province, but not with DN. The G allele may confer susceptibility to the development of T2DM patients.
