Objective:To investigate the correlation between single nucleotide polymorphisms(SNP45 and SNP276) of adiponectin gene and type 2 diabetes in XinJiang Uigur Population. Method: A 1:1 Matched Case-control Study was conducted with 120 cases of type 2 diabetes and 120 population-based controls in XinJiang Uigur Population.Adiponectin gene polymorphisms genotypes were detected by PCR-RFLP. The serum adiponectin concentration was measured by ELSA in all of subject.The relationship between the heredity porlymorphism of Apm-1 and type 2 diabetes susceptibility, the date was analysed with statistics software of the SPSS 11.0 , The odd ratios (OR) and the 95% confidence interval (CI) represent the relative risk. We analyse the 240 samples'gene porlymorphism (including 120 normals and 120 patients) and discuss of the Uigur Population. At last we analyse the data and verify the samples'population representativeness with the Hardy—Weinberg equilibration law. Theχ2 test was adopted to check the difference in genotype distribution between patients and controls.Result: (1) The serum adiponectin concentration in control group was between 0.04 and 31.79μg/mL,the average value was(6.89±5.30)μg/mL ,it was 6.911±5.79μg/mL in male group, 8.935±4.18μg/mL in female group. Serum adiponectin concentration in women was significantly higher than that of men(P<0.05) . The serum adiponectin concentration was negatively correlated with Body Mass Index (BMI), Low density lipoprotein-C (LDL-C ) and FINS. (2)The serum adiponectin concentration in disease group was between 0.40 and 18.07μg/mL,the average value was (5.48±4.04)μg/mL; Serum adiponectin concentration in control group was significantly higher than that of disease group (P=0.0000) . There is significant difference in the blood fat concentration between disease group and control group (P<0.05). (3) The distribution of genotype of SNP45 in case/control group was as follows: TT 67/60(55.83%/50%);GT 36/45(30%/37.5%),GG 17/15(14.17%/12.5%);The distribution of the T and G allele in case/control group was as follows: T allele 170/165(70.83%/68.75%);G allele 70/75(29.17%/31.25%).The frequencies of the SNP45 polymorphism were not different in diabetes and control groups(P>0.05).(4) The distribution of genotype of SNP276 in case/control group was as follows: GG 83/66(69.17%/55%);GT 29/45(24.17%/37.5%),TT 8/9(6.7%/7.5%);The distribution of the G and T allele in case/control group was as follows: G allele 195/177(81.25%/73.75%);T allele 45/63(18.75%/26.25%). The distribution of genotype frequencies of the SNP276 polymorphism were not different in diabetes and control groups(P>0.05), but the allele frequencies has significant difference in diabetes and control groups(P=0.049).(5) The serum adiponectin concentrations of these three genotypes of SNP45 were as follows : In disease group, TT(8.53±4.89μg/mL),GT(3.74±2.51μg/mL),GG(4.83±3.73μg/mL);In control group , TT(15.8±6.30μg/mL),GT(6.76±4.46μg/mL),GG(5.36±3.153μg/mL), there were significant differences among these three genotypes (P=0.0000). The genotype TT is higher than GG and GT in the serum adiponectin concentrations.(6) The serum adiponectin concentrations of these three genotypes of SNP276 were as follows : In disease group, TT(11.4±5.78μg/mL),GT(6.79±4.68μg/mL),GG(3.80±2.23μg/mL);In control group , TT(18.8±5.96μg/mL),GT(8.45±4.03μg/mL),GG(4.75±3.22μg/mL),there were significant differences among these three genotypes (P=0.0000). The genotype TT is higher than GG and GT in the serum adiponectin concentrations.Conclusion:(1) The results suggest that in the normal person of Xinjiang Uigur population, the serum adiponectin concentration in women was significantly higher than that of men(P<0.05). The serum adiponectin concentration was negatively correlated with Body Mass Index (BMI), Low density lipoprotein-C (LDL-C ) and FINS. (2)The blood fat metabolic disorder was dangerous factor of type 2 diabetes in XinJiang Uigur Population ;The serum adiponectin concentration was protection factor of type 2 diabetes in XinJiang Uigur Population .(3)The SNP45 in adiponectin gene were not associated with type 2 diabetes in XinJiang Uigur Population. (3) The SNP276 in adiponectin gene were associated with type 2 diabetes in XinJiang Uigur Population; (4) The serum adiponectin concentration was correlated with the SNP45 and the SNP276 in adiponectin gene.
