Objective:To investigate the expression of B7-H3 and B7-H4 molecule in human ovarian serious and mucinous carcinoma. To explore the relation with B7-H3 and B7-H4 molecule in ovarian serious and mucinous carcinoma generation and development combined with the clinicopathologic datas. To provide an application prospect in ovarian epithelials cacinoma early diagnoses and the target therapy.Methods:Immunohistochemical (IHC) PV-9000 method and reverse transcrption-polymerae chain reaction(RT-PCR) method were used to detected the expression of B7-H3 and B7-H4 in groupⅠand groupⅡ, respectively. There are 75 cases of ovarian serious tumor in groupⅠ, include of 40 cases of serious carcinoma. 20 cases of serious borderlin cystadenoma and 15 cases of serious cystadenoma. There are 70 cases of ovarian mucinous tumor in groupⅡ, include of 30 cases of mucinous carcinoma,20 cases of mucinous borderlin cystadenoma and 20 cases of mucinous cystadenoma.Results:1.Immuohistochemistry experiments results.1.1 B7-H3 protein expressed on the both groupⅠandⅡ. In groupⅠ, the positive proportion of B7-H3 protein in serious cystadenoma, serious borderlin cystadenoma andserious carcinoma were 13.3%,35.0%and 85.0%,respectively. The positive expression rates increase progressively. There was significant difference between serious carcinoma and serious borderlin cystadenomaor serious cystadenoma,respectively (p<0.01).There was no statistical difference between serious borderlin cystadenomawith serious cystadenoma (p>0.05), but the positive expression rates in serious borderlin cystadenomais higher than in serious cystadenoma. In groupⅡ, the positive expression rates of B7-H3 in mucinous cystadenoma, mucinous borderlin cystadenoma and mucinous carcinoma were 0%,10.0% and 50.0%,respectively. The positive expression rates indcrease progressively. There was significant difference between mucinous carcinoma and mucinous borderlin cystadenoma or mucinous cystadenoma,respectively (p<0.01).There was no statistical difference between mucinous borderlin cystadenoma with mucinous cystadenoma, but the positive expression rates in mucinous borderlin cystadenoma is higher than in mucinous cystadenoma.1.2 B7-H4 protein expressed on the both groupⅠandⅡ. In groupⅠthe positive proportion of B7-H4 protein in serious cystadenoma, serious borderlin cystadenomaandserious carcinoma were 13.3%,50.0% and 75.0%,respectively. In group II, the positive expression rates of B7-H4 protein in mucinous cystadenoma, mucinous borderlin cystadenoma and mucinous carcinoma were 0%,20.0% and 50.0%,respectively. There was statistically difference in each two group (p<0.05).1.3 The difference was statistically significant between B7-H3 and B7-H4 in serious carcinoma and mucinous carcinoma, respectively(p<0.05).1.4 There was no correlation between B7-H3 protein expression and clinical stage, pathological grade, age, tumor size, lymph node metastasis and astites cytology(p>0.05) in serious carcinoma or in mucinous carcinoma. The same result was expressed of B7-H4 protein.1.5 There was positive correlation between the expression of B7-H3 and B7-H4 in serious carcinoma and mucinous carcinoma, respectively(p<0.05).2.RT-PCR experiments results.2.1 B7-H3 mRNA and B7-H4 mRNA were detected in both groupⅠandⅡ.2.2 In groupⅠ,B7-H3 mRNA in serious cystadenoma, serious borderlin cystadenomaandserious carcinoma indcrease progressively. There was significant difference between serious carcinoma and serious borderlin cystadenomaor serious cystadenoma,respectively (p<0.01).There was no statistical difference between serious borderlin cystadenomawith serious cystadenoma (p>0.05).In groupⅡ, B7-H3 mRNA in mucinous cystadenoma, mucinous borderlin cystadenoma and mucinous carcinoma indcrease progressively. There was significant difference between mucinous carcinoma and mucinous borderlin cystadenoma or mucinous cystadenoma,respectively (p<0.01).There was no statistical difference between mucinous borderlin cystadenoma with mucinous cystadenoma (p>0.05).2.3 In group I,B7-H4 mRNA in serious cystadenoma, serious borderlin cystadeno ma and serious carcinoma indcrease progressively. There was significant difference in each two group (p<0.01). In groupⅡ, B7-H4 mRNA in mucinous cystadenoma, mucinous borderlin cystadenoma and mucinous carcinoma indcrease progressively. There was significant difference in each two group (p<0.01).2.4 There was signficant differenct between B7-H3 mRNA and B7-H4 mRNA in serious carcinoma and mucinous carcinoma, respectively(p<0.05).2.5 There was positive correlation between B7-H3 mRNA and B7-H4 mRNA in serious carcinoma and mucinous carcinoma, respectively(p<0.01).Conclution:1. Both in protein and mRNA, the expressions of B7-H3 and B7-H4 in ovarian malignancy carcinomas were higher than in bodeline and benign cystadenomas (P<0.01) which suggest that B7-H3 and B7-H4 may be the important factors in ovarian malignancy carcinoma form,and involve in ovarian cancer generation and development.2. There was difference expressions of B7-H3 and B7-H4 between in serious carcinoma and in mucinous carcinoma (P<0.05). These two types carcinoma arised from the different origin may be the mainly reason.3. The positive correlation between B7-H3 and B7-H4 in serious carcinoma and mucinous carcinoma, respectively, which suggest that there must be a part of B7-H3 biological activity in ovarian cancer is similar with B7-H4, they may be both the inhibition factor of T cell activation to promote ovarian cancer occurrence and development. These two molecules were at least of part biological activity and probably become new targets of diagnosis and/or treatment for ovarian serious and mucinous carcinoma.
