Nepeta angustifoliathe G Y. WU pertains to the Nepeta genus of Labiatae family. It's the original Tibetan medicine plant from a area of Tibet which is called ShaDuna-bao.It grows in hillside, roadside, the lawn, the edge of forest, at the elevations between 3600 and 4600m. It's mainly distributed in the central and south areas of Tibet. Nepeta plays the roles of causing resuscitation and restoring consciousness, in the Tibetan Medicine is mainly used to treat dizziness and faint apoplexia, epilepsia, cerebral hemorrhage, cephalomeningitis, pestilence, pain, skin and external diseases, etc.In this paper, the orthogonal experimental design was used to optimize the steam distillation condition for essential oil from Nepeta Angustifoliathe G Y. WU. We confirmed the optimum conditions were the immersion time was 2h, the fluid material ratio was 10:1 and extraction time was 3h. The chemical constituents of essential oil were extracted by steam distillation from Nepeta Angustifoliathe G Y. WU and identified by GC-MS. The relative content of each constituent was determined by area normalization method.29 compounds were identified and determined. The main components and contents were nepetalactone (75.37%), spiro(5.6), dodecane (3.47%),2,4-dimethyl-1,3-pentadiene (2.70%), bisabolol (1.45%),5-nonene-2-one (1.0%),etc. such 24 compounds as dehydro-nepetalactone, niaouli cineole, bisabolol and so on were all isolated from the Labiatae family plant for the fist time.It demonstrates that the constituents of essential oil are very different from the other Nepeta Angustifoliathe.Recirculation was used to extract the whole plant of Nepeta angustifoliathe G Y. WU with alcohol, through ligarine, dichloromethane extracting and under pressure concentrating; the extraction phases including various kinds of chemical compositions were obtained. Column chromatography and recrystallization were applied to separate the extraction phase of dichloromethane. Three monomer compounds were obtained and the structures were identified by physicochemical property, thin-layer chromatography and spectroscopic method. They were betulinic acid, methyl oleanolate, and ursolic acid. All of the compounds were gained from Nepeta angustifoliathe G Y. WU for the first time.The ultraviolet spectrophotometry was established to determine the content of total flavone in Nepeta angustifoliathe G Y. WU. Rutin was used as reference substance, and sodium nitrite-aluminium nitrate was used as colour-developing agent. The result shows that the method is easy, fast and accurate. The average returns-ratio is 99.07%, RSD is 0.87%. The total flavanone content determined by this method is 7.71mg/g.This law can be taken as content determination method in the total flavanone extraction process research, At the same time this research wil provid examples of experiences and theoretical basis for the determination of total flavonoids in other drugs.In this paper, the content of total flavone was taken as the evaluating indicator, the influence to extraction factors such as ethyl alcohol concentration, the liquid-solid rate, the extraction time were investigated. Through L9 (33) the orthogonal experimental design, we confirmed the optimum conditions was 70% ethyl alcohol, the ratio of liquid to solid was 16:1, extraction time was 45min.The extraction of total flavone from Nepeta angustifoliathe G Y. WU by supercritical carbon dioxide extraction was studied. Taking the content of total flavone as the evaluating indicator, the influence to extraction factors such as the extraction temperature, the extraction pressure and the extraction time were investigated. By L9 (33) the orthogonal experimental design, we confirmed the optimum conditions were as follows:extraction temperature, extraction pressure and extraction time were 40℃,26 MPa, 1h respectively.
