Objective: 1.To establish a new method of quality evaluation and validate its feasibilities by the simultaneous quantitative assay of four components in Coreopsis tinctoria Nutt.2.To build laboratory and pilot extraction processing of Coreopsis tinctoria ethyl acetate extracts.3.To develop an HPLC-MS method for identifying the chemical compounds in Coreopsis tinctoria Herbs and Its Extracts.4.To conduct preliminary study of extract of coreopsis tinctoria intestinal absorption and blood transitional components.Methods: 1.A HPLC method was developed as QAMS to determine Flavanomarein、Marein and Dicaffeoylquinic acid in Coreopsis tinctoria Herbs and Its Extracts.2.Build chemical compounds identification method of Herbs and Extracts of coreopsis tinctoria through HPLC-MS.3.The isolated everted intestine model was used to study the absorption of flavonoid extract of coreopsis tinctoria in rat intestin.4.Study constituents absorbed into blood of flavonoid extract of coreopsis tinctoria through TCM serum pharmacochemistry.Result: 1.The relative correction factor(RCF)of other three components using Chlorogenic acid as the internal reference substance,were 0.669,3.595,0.759.And no significant differences were found in the quantitative results of Flavanomarein、Marein and Dicaffeoylquinic acid in different batch samples of Coreopsis tinctoria determined by external standard method and QAMS(P﹥0.05).2.Identificate 11 kinds of chemical compounds in extracts of coreopsis tinctoria through HPLC-MS.3.There were 13 compositions were detected in alcohol extract and ethyl acetate extract,11 compositions were detected in water extract and extraction tailings could be absorbed directly into intestinal;4.One compositions of alcohol extract and ethyl acetate extract could be absorbed into blood;Three compositions of water extract could be absorbed into blood.Conclusion: 1.It is feasible and suitable to evaluate the quality of Coreopsis tinctoria Nutt by QAMS.2.There were 11 compositions were detected in flavonoid extract of coreopsis tinctoria.3.We could infer the metabolic processes of chemicals by analyzing and comparing HPLC-MS fingerprint spectrum of serum samples and the sample after intestinal metabolism;Three compositions could be absorbed into blood.
