The Factors Related To The Depression In Rat Model Of Parkinson's Disease

Objective:Study the abnomal behaviouristics of the rats and the expression of 5-Hydroxytryptamine (5-HT) of brain, to analyze the pathogenesis and correlative factor of the depression in Parkinson disease.Methods:Male and adult Wista rats(weight 260-280g),30 rats were randomly divided into 2 groups for control and operated group. Establish a rat model of Parkinson disease, take 45 of them, randomly divided into 3 groups,and separately gave them NS, Madopar and artane.The control group and operated group were given NS too. After 3months,we observe the behaviour and the consumption test of sucrose solution,to evaluate the depression of the rats.Detect the expression of 5-HT by immunohistochemistryResults:1 Behaviouristics:In control and operated groups. the rats were runningm,looking about and smelling more than the PD rats.2 Weight and consumption test of sucrose solution:The PD group and the artane group and madopa group weight lighter than, the artane and madopa groups weight lighter than PD group,but P>0.05. The consumption of sucrose solution in the PD group and the artane group and madopa group is lower than that in control and operated groups(P<0.05).And in artane and madopa group,the consumption of the both latter group were lower than that in PD group(P<0.05)3 Immunohistochemistry The average count value of 5-HT of Hippocampus and frontal lobe in madopa group is less than that in PD group, artane group, control and operated group(p<0.05). The average of count value of 5-HT of Hippocampus and frontal lobe in artane group and PD group is lower than that in t in control and operated group but p>0.05Conclusion:1 By observe the behaviouristics of the rats and measure the weight, we found that depression exist in all the rat of Parkinson disease appear2 The artane and madopa group had worse symptom than PD group, which indicated that medicine treatment by artane and madopa can aggravate the depression in PD patients.3The expression of 5-HT is lower in madopa group,promoted that the injury of the 5-HT pathway may play an important role in depression of PD patients treated by madopa.4 Artane can induce or aggravate depression in PD, which maybe related to it's impact in cognitive impairment.It needs further study to conform.
