Objective To investigate the association between the tryptophan hydroxylase-2 (TPH-2) G1463A,monoamine oxidase A(MAOA), 5-Hydroxytryptamine 1A receptor(5-HT1A) C-1019G and 5-Hydroxytryptamine 2A receptor(5-HT2A) His452Tyr genes polymorphisms and unipolar depression in a Chinese Han population.Methods The allelic and genotypic frequencies of TPH2 G1463A, MAOA, 5-HT1A C-1019G and 5-HT2A His452Tyr genes polymorphisms were examined with techniques of polymerase chain reaction(PCR), amplification refractory mutation system-PCR(ARMS-PCR) and restriction fragment length polymorphism (RFLP) and analyzed statistically in 123 unipolar depressive patients and 122 healthy individuals.Results (1)No subjects among the cases or controls were found to carry TPH2 1463A mutant allele. All subjects were TPH2 G1463G homozygotes. (2)There were no significant differences between cases and controls in genotypic and allelic frequencies of MAOA. (3) There were no significant differences between cases and controls in genotypic and allelic frequencies of 5-HT1A C-1019G. (4) There were significant differences between cases and controls in genotypic and allelic frequencies of 5-HT2A His452Tyr. The frequencies of Tyr/Tyr genotype and Tyr allele in case group were significantly higher than those in control group. However, the difference between cases and controls was significant only in female subjects but not in male subjects after sample stratification by sex. In patient group, the frequencies of Tyr /Tyr genotype and Tyr allele in patients without suicidal ideation were significantly higher than in those with suicidal ideation.Conclusions (1)There may be no association between the TPH2 G1463A polymorphism and unipolar depression in the Chinese Han population. (2) There may be no association between the MAOA polymorphism and unipolar depression in the Chinese Han population. (3) There may be no association between the 5-HT1A C-1019G polymorphism and unipolar depression in the Chinese Han population. (4) The polymorphism of 5-HT2A His452Tyr may be related with depression, and 452Tyr allele may be a risk factor for depressive patients, especially for females.
