Traditional Chinese medicine is a complex system of diverse components. We learn from the international pharmaceutical industry of "comparability of evaluation of herbal medicine" technology system, and proposes:herbal extracts can be divided into the basic active substance and associated materials. Its therapeutic properties is completely or largely associated with the basic active substance; associated material can be divided into additional substances and non-active by-products. Additional substances can enhance or diminish the role of the basic active substance; non-active by-products have no pharmacological effect, and the existence is often not desired. To the same study system, using different purification methods will leads to different retention rate of basic active substances and different removal rate of the associated extracts; using same purification of different parameters will lead to different retention rate of basic active substances and different removal rate of the associated extracts too. The latter can play a greater role on the explanation of the effects of the associated substance.This thesis is to study the system of huanglianjiedu decoction. Ultrafiltration method in different relative MWCO pes membrane were used to got a series of samples. Physical and chemical methods were used to measure the content of the efficacy substance and the physical and chemical parameters in the refined sample. In vitro experimental, Refined samples were added to PC 12 cells to compare the protective effects on ischemia and hypoxia. In this thesis, with industrial prospects of membrane separation technology, get new ideas to study the material basis of traditional Chinese medicine, thereby establish reasonable evaluation system for separating and refining traditional Chinese medicine techniques (especially on membrane technology).Content and the Result:(1) The solids fraction of the UF extract were determined according to the method of Pharmacopoeia of the People's Republic of China (2005 edition). The solid fraction of the centrifugal liquid,pes-5w, pes-3w, pes-2w, pes-lw and pes-6k were 19.451,16.884,16.313, 15.040,13.534,12.894.(2) The physicochemical properties of the UF extract were carefully determined. pH of the centrifugal liquid, pes-5w, pes-3w, pes-2w, pes-lw and pes-6k were 4.002,4.278,4.337,4.431, 4.500,4.743. Conductivity of the centrifugal liquid,pes-5w, pes-3w, pes-2w, pes-lw and pes-6k were 1.943,2.540,2.630,2.800,3.040,3.740. Turbidity of the centrifugal liquid,pes-5w,pes-3w, pes-2w, pes-lw and pes-6k were 288.0,73.2,56.9,47.1,40.2,10.9 Viscosity is notregular.it is at about 8.0. The content of the protein, pectin and starch diclines in the UF process.(3) HPLC was used to determine the contents of berberine, gardenoside, baicalin and palmatine of the UF extract. This method shows good sensitivity, high selectivity and good separation. The content of the gardenoside of the centrifugal liquid, pes-5w, pes-3w, pes-2w, pes-lw and pes-6k were 0.463,0.518,0.511,0.522,0.560,0.622 mg/mL.The content of the baicalin of the centrifugal liquid, pes-5w, pes-3w, pes-2w, pes-lw and pes-6k were 0.468,0.336, 0.324,0.263,0.260,0.235 mg/mL.The content of the palmatine of the centrifugal liquid, pes-5w, pes-3w, pes-2w, pes-lw and pes-6k were 0.194,0.201,0.198,0.200,0.202,0.212 mg/mL. The content of the berberine of the centrifugal liquid, pes-5w, pes-3w, pes-2w, pes-lw and pes-6k were 0.789,0.693,0.704,0.622,0.630,0.618 mg/mL.(4) HPLC was used to compare the characteristic spectrum of the UF extract. Chemical composition and the content changes were compared.(5) Carry dissolution experiment and the content of berberine, gardenoside, baicalin and palmatine were determined. There is no common among these four efficacy substance.(6) Damage the PC 12 cell, measure the rate of protection of all the samples. The protection of the pes-5w UF extract is the best.The innovation of this thesis:(1) Use the turbidity to study the huanglianjiedu decoction for the first time.Turbidity is used to describe the state of the microparticle.The temperature of the experiment is confirmed.(2) Use 5 different relative molecular weight cutoff of membrant to study the huanglianjiedu decoction for the first time. (3) Characteristic spectrum is use to the comparison between the the centrifugal liquid and the UF extract.(4) Determine the dissolution of the huanglianjiedu decoction which is centrifugated or ultrafiltrated.The dissolution of berberine, gardenoside, baicalin and palmatine is different.
