Using Dangguibuxue decoction as a model drug, the project studied preparation technique of Dangguibuxue decoction by orthogonal test and the optimal were as follows:the dried herbs of Dangguibuxue decoction were boiled three times with twelve times of water for one hour by orthogonal design. According to the properties of liquid disperse system, the phase state of Dangguibuxue decoction was studied too, which was composed of the true solution in homogeneous state, the colloidal solution in heterogeneous state, the suspension in heterogeneous state or the emulsion in heterogeneous state.On this basis, referring to research methods of different dispersion system, using water as dispersion medium, physicochemical characterization of Dangguibuxue decotion and its single medicines and main active ingredients, such as astragaloside Ⅳ, ferulic acid, astragaloside, polysaccharides were studied, including the surface properties, electrical properties and rheological property. It was found that the surface tension was mainly affected by astragaloside IV, both the pH value and the conductivity were mainly affected by ferulic acid, and the osmotic pressure might be mainly affected by polysaccharides. Surface tension, conductivity and osmotic pressure could reflect the quality of Dangguibuxue decoction.In addition,under the effect of invigorationg Qi and enriching blood of Dangguibuxue decoction, aiming at blood deficiency model mice induced by injecting cyclophosphamide (CY) and qi deficiency model mice induced by compound factors, relationships among physicochemical parameters and component content and the effect of Dangguibuxue decoction were studied too. It was found that the Surface tension, conductivity and osmotic pressure could reflect the effect of invigorationg Qi and enriching blood of Dangguibuxue decoction.At the same time, through polynomial regression analysis, results showed that physicochemical parameters closely correlated with component content and effect of invigorating Qi and enriching blood of Dangguibuxue decoction. These results preliminarily suggested that physicochemical parameters could be used to characterize the effect and quality of decoction.
