Research of compound of Chinese materia medica based on separating and refining techniques is common method that modernization research of medicine compound to be faced with. Refining process based on the separation technique is the key method of developing from simple decoction compound to modern formulations. Therefore, this dissertation mainly analyzes the rationality of separating compound of Chinese materia medica by means of different refining techniques, to explore and build modern separation and purification theories and technological platform of compound of Chinese materia medica based on industrialization separation methods.This dissertation focused on Coptidis Decoction for Detoxification——the classic formula of heat-clearing and detoxicating of Chinese medicine, as a research object. We applied some techniques such as macroporous adsorptive resins, membrane separation, extracted by water and then precipitated by alcohol, flocculation to prepare different combinations of samples. And we apply pharmacokinetics to measure the different physiological disposition of baicalin among the samples in rats by intragastric administration. Elucidate the material foundation of the different refining techniques of Coptidis Decoction for Detoxification, and build a rational evaluation of different separating and refining technology of compound of Chinese materia medica.Physical pharmacy and biological pharmacy are foundation of dosage form, preparation design and preparing craft. To research the differences of composition among the compound by different refined crafts in physical pharmacy and biological pharmacy, the relationship between the physicochemical properties of samples and absorption of effective ingredient in vivo in rats is detected. Overview of this dissertation as follows: Objective: we applied a variety of modern separating and refining technology, to search for new ideas and new methods for studying on the material foundation of formula of Traditional Chinese medicine, and intended to establish the separation theory and technology system that have industrial prospect compound.Content:1. Research of the optimal crafts and refinements of water extractives of Coptidis Decoction for Detoxification.2. Research of the diversifying rules of solids fraction among the different refined products.3. Research of the diversifying rules of physicochemical properties among the different refined products;4. Research of the diversifying rules of alkaloid contents among the different refined products;5. Research of the diversifying rules of jasminoidin, baicalin and berberine hydrochloride among the different refined products;6. Research of the diversifying rules of the quantities of baicalin in the plasma among the different refined products;7. Research of the diversifying rules of the platelet aggregation functions among the different refined products;Method:1. To determine the optimal crafts and refinements of water extractives of Coptidis Decoction for Detoxification, UV spectrophotometry was used.2. The solids fraction of the different refined products were determined according to the method of Pharmacopoeia of the People’s Republic of China (2005 edition).3. The physicochemical properties of the different refined products were carefully determined.4. UV spectrophotometry was used to determine the alkaloid contents of the different refined products;5. HPLC was used to determine the contents of jasminoidin, baicalin and berberine hydrochloride of the different refined products;6. The mice were divided into groups in random and perfused with all the refined products extracted from Coptidis Decoction for Detoxification. At the points of 0.0833, 0.167, 0.333, 0.5, 1, 1.5, 2, 3, 4, 6, 8, 10, 12, 16, 24h, picked off the eyeballs of the corresponding mice to get blood samples for determining the quantity of baicalin in the plasma. Concentration-Time Curves of Baicalin in mice’s Blood was drew properly.7. The pharmacokinetics parameters of the different refined products were Calculated8. Platelet aggregation rate within 5 minutes was measured by turbidimetric method with Instrument of STEELIEX Platelet Aggregation Coagulation Factor Analysis, recorded the largest aggregation, and calculate platelet aggregation inhibition rate.Results:1. Use of HPLC to determine Berberine, Baicalin and jasminoidin under the same condition is established. It’s simple, accurate, sensitive and stable method.2. Use of UV spectrophotometry to determine amount of flavonoids and alkaloid is simple, reliable and accurate method.3. Use of HPLC is valid method to determine baicalin in Rats’ Blood.4. Amount of Active constituent refined with AB-8 resin is much more than any other method.5. AUC of Baicalin in blood of rats, of which sample refined with AB-8 resin is larger than other refined crafts, which indicates that other ingredients in the sample can promote absorption of Baicalin.6. Rats perfused with all refined samples of Coptidis Decoction for Detoxification shows double-kurtosis of the concentration-time curves of baicalin.7. There is some relevance between physicochemical properties to the absorption of Berberine;8. Inhibition effect of the refined products with AB-8 resin on ADP-Induced Platelet Aggregation Rate is better than other refined products.
