Huanglianjiedu decoction is the sutra recipe of defevering and detoxifying. It consists of four medicines which are coptis root(9g), baikal skullcap(6g), corktree bark(6g), capejastmine(9g). The effective fractions of Huanglianjiedu Decoction are found to be able to inhibit 11 kinds viruses ^specially herpes virus I * II. So it is confirmed the effects of Huanglianjiedu Decoction. The dissertation divides into three parts to study the material and mechanism of anti-herpes virus of Huanglianjiedu Decoction.Part one: Literatures and summarizations about Huanglianjiedu Decoction.It sums respectively up the pharmacy, pharmacology and chemical components of Huanglianjiedu Decoction and single medicines. 70 references are cited.Part two: Experiment studies about Huanglianjiedu Decoction.One, the effects of anti-herpes virus from Huanglianjiedu Decoction are extracted and separated. 13 compounds are isolated with silica gel, polyamide, sephadex gel chromatography and 11 compounds are identified with UV. IR, EI-MS* FAB-MS?HR-MS, 'HNMR, 13CNMR spectra. They are three alkaloids (berberine,palmatine and jatrorrhizine),three flavonoids(baicalin, luteolin and wogonin),three organic acids(chlorogenic, ursolic acid and crocetin),one iridoid glycoside(geniposide) and one sterol( P -sitosterol). All of the 11 compounds are firstly obtained from Huanglianjiedu Decoction. Especially luteolin among them is firstly acquired from Huanglianjiedu Decoction, and no literatures was reported to have acquired it from the four medicines of the compound recipe.Two, HPLC figs of the effects of anti-herpes virus from Huanglianjiedu Decoction are established primarily. By analyzing and comparing, 28 chromatographic peaks are found in the HPLC figs of the effects of anti-herpes virus from Huanglianjiedu Decoction and the ascriptions of the peaks are validated in the HPLC figs of every single medicine. Some peaks are confirmed after comparing with standard samples.Three, the total alkaloids contents in the effects of anti-herpes virus from Huanglianjiedu Decoction and coptis root, corktree bark are determined. They are 13.89%, 51.34%, 9.82% respectively. The linear range of berberine is 0.0040-0.0200mg/mL, the correlation coefficient is 0.9997. the contents of index compounds including baicalin and geniposide are determined by HPLC. The calibration curves are linear at ranges of 0.572-1.716 M g and 2.02-18.18 n g respectively, the correlation coefficients are 0.9995 and 0.9992. The contents are 13.31% and 9.96% respectively, the average recoveries 102.5%(RSD1.51%,n=3)and 99.2%(RSD1.94%,n=3).Part three: analysis and discussion of experiments.
