Objective:To observe the effects of acupuncture at different acupoints on the content of PKC in the stroked rats with hemiplegia spasticity, so that explore the mechanism for acupuncture at acupoints against cerebral ischemia with hemiplegia spasticity which will provide an theory basis for acupuncture treatment and ease muscle paralysis increased tension. Methods:Using modified thread bolt for preparation of MCAO model rats, and after 3 hour of reperfusion the level of nerves'action were measured by Zea Longa 4 branch of criterion. The successful model rats were started treatment after the third day, and continuoued treatment for 14 days, once per day. Using neural physiological H-reflex evaluation model and treatment effect. And the protein expression of PKC which were exist in brain and spinal cord of rats were detected by western-blot. And the serum level of PKC-activated which were exist in blood serum of rats by using enzyme-linked immunosorbent assay (Elisa)Results:1,the right H wave latency, yang meridians acupuncture group than in model group was significantly longer, and was significantly different with the model group, Yin meridians acupuncture, yin and yang meridians by the acupuncture group, baclofen group, MCPG groups (P<0.01, P<0.05); the right side of H-reflex recovery curve showed yang meridians acupuncture group H/M values up to 1.078±0.1283, and was significantly different with the model group, Yin meridians and acupuncture, yin and yang meridians by the acupuncture group, baclofen group, MCPG group (P<0.05, P<0.05); 2,Yang meridians acupuncture group were significantly lower in the serum level of PKC-activated in rats'blood serum and the protein expression of PKC in rats'brain and spinal cord, and the model group, Yin meridians and acupuncture, yin and yang meridians by acupuncture group, baclofen group, MCPG group were significantly different whit it.(P<0.05, P<0.05).Conclusion:1,Acupuncture significantly inhibited hyperthyroidism of H-reflex and improve the H reflection restore curve, restrain the totor neuronal excitability of spinal cord, alleviate the rats'hemiplegia spasticity; 2,acupuncture treatment reduce the content of PKC in brain and spinal cord and the content of PKC-activated blood serum of the stroked rats with hemiplegia spasticity, three groups of acupuncture group were different in statistics;3,Acupuncture treatment alleviate hemiplegia spasticity after stroke. Three groups of acupuncture have differences in statistics, acupunctured in Yang meridians has the strongest function, acupunctured in Yang and Yin meridians comes after, but in Yin meridians is the weakest.
