Objective:To discuss the effects of bronchial asthma in mice exposed to different concentrations of FluorantheneMethods:70 male Balb/c mice were randomly divided into seven groups (every group=10). The mice in C group, which was a asthma model group, were given intraperitoneal injections to Ovalbumin(OVA) 20ug+Al(OH)3 lmg, total 2ml, at the In 8,15 day and given 3%OVA+saline inhalation for 30minutes on conescutive seven days since 22th day. The mice in A group, which was a control group, were given intraperitoneal injections and inhaled to normal saline insteading of OVA, step with C group. Group B was Alcohol control group, the mice of B group were intraperitoneal injected and inhaled with 95%ethanol insteading of OVA, step with C group. Group D was low concentration (10ug/m3) of fluoranthene group. Group E was high concentration (100ug/m3) of fluoranthene group. Group F was low concentration of fluoranthene and asthma model group. Group G was high concentration of fluoranthene and asthma model group. The mice of D, E, F, G groups were exposed to the different concentrations of fluoranthene in 2 weeks,4h/d. The mice of A, B, C groups were exposed to fresh air at the same time. F, G groups after exposure to exposure, then sensitized to OVA, step with C group. A week later, all groups were sensitized with 3%OVA for 30 minutes, then test the change of airway resistance were observed for 15 min and counted eosinophils in bronchial lavage fluid and detected the content of MDA, SOD and GSH-Px in lung tissue and measured the expression of serum immunoglobulin E (IgE), interleukin-4 (IL-4) and interleukin-5 (IL-5) byenzyme-linked immunosorbent assay (ELISA). Finally observe the pathological changes of lung tissue.Results:①After inhalation of MCH, the ratio of airway resistance in A, B groups were similar. Compared with the control group, the ratio of airway resistance in C, D, E, F, G groups were significantly increased. Differences of C, F, G groups were statistically significant (P<0.01). Compared with the C group, the ratios of airway resistance of F, G groups had increased, only G group was statistically significant (P<0.01).②In bronchial lavage fluid of A,B group there were a small number of eosinophils (EOS). EOS of C, D, E groups accounted for a percentage of total cells were significantly higher than the control group, the differences were statistically significant (F=55.84, P<001). Compared with the C group, EOS of D,E groups accounted for a percentage of total cells had increased, but no statistical differences (P> 0.05).③The content of MDA, SOD and GSH-Px in A, B, C groups were almost similar, they had no significant differences. The content of SOD and GSH-Px in group D, E, F, G were lower than the control group; and the content of MDA were higher than the control group, and they had significant differences (P< 0.01). In E, G groups, the content of SOD and GSH-Px were lower than D,F group; and the content of MDA were higher than D,F group. They had significant differences (P<0.01).④The expression of IgE, IL-4 and IL-5 in A, B, D, E groups were almost similar. Compared with the control group, the expression of IgE,IL-4 and IL-5in C, F, Ggroup had significantly increased, and they had significant differences (P<0.01). Compared with C group, the expression of IgE, IL-4 and IL-5 in F, G groups had increased, but had no significant differences (P> 0.05).⑤The lung pathological section showed that the endothelial cells in A, B group were integrated, and had no denaturation or necrosis, stroma did not see of inflammation cell infiltrate, periviascular did not see edema. In C, D, E, F, G groups we appeared that epithelial cells had different levels necrosis, interruption, fall off, the airway inflammatory cells infiltration and perivascular edema. And the higher fluoranthene concentration, lung tissue damaged degree is heavier.Conclusion:Exposed to Fluoranthene in the mice can enhance lipid peroxidation and increase lung infiltration of inflammatory cells. Exposed to high concentrations of Fluoranthene can aggravate the condition of asthmatic mice.
