Objective:This papers in medicine "six pathogens disease"as the theoretical basis, through the preparation of alpine environments and upper respiratory infections in mice have a study on non-biological pathogenic factors - cold dampness pathogen and biological virulence factors (virus) on immune function. Dampness pathogen to the pathogenesis of upper respiratory tract infection caused by the awareness and prevention of disease alpine provide experimental basis.Methods:1 Group dispensation:96 female Kunming mice, weighing 30±2g, were randomly divided into 8 groups, namely:normal group,virus group,Chinese medicine virus group,Western medicine virus group,Chinese medicine cold-damp group,Western medicine cold-damp group.Each group contains 12 mice.2 Modeling and administration:(1)Preparing upper respiratory tract infection model:The normal mice were normally bred 2 days, and then given the mice upper Revised Standard Version (RSV) 106PFU 0.1ml per mice through their noses except the normal group and the cold-damp balance group.(2)Given the Chinese medicine model group Ma Huang Jia Zhu Tang and the Western medicine model group virazole by pouring the medicine into stomach for 7days.(3) The use of artificial climate chamber set up alpine environment model, the Alpine group, cold + virus infection, cold + virus Medicines, cold + virus infected mice placed in western alpine environment model, Every day to stimulate the 8h, for 7 days.3 Detection:(1)Observed in normal mice and the alpine environment, changes in general, the mice mental state, activity, fur, skin, joints, nose, and the shit and pee of the mice, take down the food and water the mice take in and their body weight in regular time.(2)The use of respiratory syncytial virus upper respiratory tract infection in mice, placed for 7 days after the drawn alpine environment. There are specific blood eyeballs mice, centrifuged and detection of immune targets in serum (serum IL-2, IFN-y content); The spleens of mice were taken aseptically, and then the ability of lymphocyte proliferation in spleen was tested with the method of MTT.Results:1 Compared with normal mice, mice in each model are generally changed during the whole modeling the activities of the model group decreased significantly, appears apathetic, slow growth or reduce body weight, eating, drinking reduced, there an increase in nasal secretions, stool thin, dull coat, some mice appeared swollen joints, and even ulceration and so on.2 The model group serum IL-2, IFN-y levels, lymphocyte proliferation was significantly lower than the.normal group, the statistical treatment, there were significant differences (P<0.05 or P<0.01). Viral infection in which the decline in alpine is far more significant. Serum IL-2, IFN-y levels and spleen lymphocyte proliferation experiments showed that alpine + infection group compared with the virus in the alpine group was obvious.3 Western medicine group medicine group and the group of mice than viruses, cold group, cold + virus infection improved mental status, weight gain, joint normal, decreased nasal secretions, stool forming; and serum IL-2, IFN-γlevels and spleen lymphocyte proliferation than the virus group, cold group and alpine virus group, indicating that the drug intervention can influence the development of disease.Conclusions:By exploring the pathogenesis of upper respiratory tract infection, the establishment of alpine environments, and upper respiratory tract infection in normal mice health status, immune status, through the influence of drug intervention, the following conclusions:1 And different from normal mice, cold environment and the general condition of mice infected with different degrees of change, the general state of poor, damp pathogen with the virus show will affect the activities of mice, eating, drinking, body weight and so on.2 Alpine environment can reduce the normal and the upper respiratory tract infection in mice serum IL-2, IFN-7 levels, leading to low immunity; and upper respiratory tract infection in mice serum IL-2, IFN-γlevels decreased more shows that alpine environment, upper respiratory tract infection in mice can affect the immune status of normal alpine environment on the same environment and the immune status of mice have different effects.3 Alpine environment can make normal and upper respiratory tract infection lymphocyte decline in the value-added, which reduces the immunity of mice; alpine environment of the upper respiratory tract infection in mouse spleen lymphocytes in a significant drop of value-added,The alpine environment on the incidence of upper respiratory tract infection and development of mice has a direct influence, that cold-damp + virus in the pathogenesis of upper respiratory tract infection was significantly enhance the pathogenesis of toxicity.4 Upper respiratory tract infection in mice and alpine environment, upper respiratory tract infection in mice, drugs in the intervention of Western medicine, Chinese medicine and western medicine group general mental status improved; mice serum IL-2, IFN-γContent, value-added lymphocytes higher than the virus group, cold group and alpine virus group, indicating that Western medicine can interfere with the disease. As indicators of the restrictions observed in this experiment, Chinese and Western medicine in the intervention there was no difference in the disease.
