ObjectiveObserve the influence of Erbao Granules on hormone VIP, MTL, Ghrelin and downstream signaling pathways and further clarify the mechanism of gastrointestinal motility disorder and the therapeutic mechanism of Promoting Digestion and Appetite to provide experimental and theoretical basis for clinical application.MethodsErbao Granules-contained serum effect on antral muscle cells to discover the expression of gastrointestinal peptides and relevant receptor, and then observe the Erbao Granules'regulation to VIP, MTL and Ghrelin signaling pathways, and finally to define whether there are collaborative or antagonistic relations among VIP, MTL and Ghrelin after the Erbao Granules treatment.ResultsThe quantitative PCR reveals that 24 hours after Erbao Granules-contained serum's effect on the receptor, the expression of VIP and its receptor 1 and 2 and Ghrelin mRNA both increase, mRNA expression of Ghrelin's receptor GHSR appears upward trend, while there's no obvious change on the mRNA expression of MTL. The Western blotting reveals that 48 hours after Erbao Granules-contained serum's effect, the expression of VIP receptor 1 significantly increase.ConclusionsBeing treated by Erbao Granules-containd serum, the expression of VIP and its receptor and Ghrelin collaboratively increase, which explains that Erbao Granules can regulate VIP, MTL and Ghrelin signaling pathways, improve gastrointestinal motility and repair gastrointestinal function, accordingly achieve the purpose of Juvenile Anorexia treatment.
