Objective:1,This subject aims to observe the effect of Astragalus infection (AI)on the vascular endothelial function, cardiac function and the clinical symptoms for the patients with acute myocardial infarction(AMI):2,To research the possible mechanism for stimulating angiogenesis of AIMethods:First, the clinical research:The patients with acute myocardial infarction were collected from Jiangsu Province hospital of Chinese medicine who were diagnosed clearly and accepted the treatments during December,2009 to December,2010. They were randomly divided into the AI group (20 cases) and the control group(20 cases). The latter was treated with conventional treatments, while the former was treated with Astragalus infection besides conventional treatments. The two groups did't show significant differences in the following aspects:age, sex, body weight index, dangerous factors, cardiac function and so on. The treatment course experienced 12±2 days. Before and after the treatment, we recorded the values of NO, ET, SOD, the graduation of cardiac function, and evaluated scores of Chinese medcine symptoms.Second, Experimental study:using in vitro human umbilical vein endothelial cells (HUMC), observation of astragalus armour vascular endothelial cells migration, proliferation influence. The datas were analyzed by SPSS 16.0, and P< 0.05 has significant difference. Results:lst, the clinical research:(1) After the treatment, all over of the patients were better than before accroding to the integral table of Chinese medcine symptoms, the improvement of the AI group was more obvious than the control group (P<0.05).(2) According to the integral table of TCM symptoms.the patients of the two groups were better than before, the improvement of the AI group was more obvious than the control group in the following aspects(P<0.01或P<0.05):languid, spontaneous sweating and the total scores.(3) the graduation of cardiac function. the improvement of the AI group was obvious and the difference had statistical significance (P<0.05). the control group improved than before, but didn't have statistical significance (P>0.05).(4) NO, ET, SOD:after the treatment,NO, SOD, ET of the patients in the AI group were better than before, the differences had statistical significance (P<0.01或P<0.05), while the control group dic't show obvious improvement; Compared with the two groups, the differences of NO,SOD,ET had statistical significance (P<0.01或P<0.05). 2nd, the experimental study:(1)the through cell proliferation experiments, we found a different concentrations astragalus quercetin (50μg/ml,100μg/ml,200μg/ml) group were able to promote HUMC proliferation role, and not dosing group compared with significant difference (P< 0.05). Between different dosage group with the increase of the concentration of the mangiferin astragalus armour, the promoting function, but also enhance the proliferation of different between no significant differences.(2)cell migration experimental results show that different concentrations astragalus armour quercetin (50μg/ml,100μg/ml,200μg/ml) group were able to promote HUMC migration, and not dosing group compared with significant difference(P< 0.05).Conclusions:(1) The Astragalus infection can improve the clinical symptoms of the patients with AMI effectively, especially on the symptoms of Qi deficiency.(2) The Astragalus infection is effective for the improvement of the cardiac function of the patients with AMI.(3) The Astragalus infection can improve the function of vascular endothelial cell, eliminate the ability of cells'oxidative stress.(4) Astragaloside can promote can promote HUMC migration, the proliferation role.
