Background and ObjectiveIt has been found that kriippel like factor 6 (KLF6) and ww domain-containing oxidoreductase (WWOX) gene are two tumor suppressor genes in recent years. Although it has been showed that the low or loss expression of KLF6 and WWOX are closely related to the development of many human malignant tumors respectively, there is no related report of the combined detection of both expression in colorectal carcinoma (CRC) and their correlation at home and abroad. And the mechanism of KLF6 and WWOX in the formation of CRC is not fully understood.The Objective of this study is to detect the expression of KLF6 and WWOX protein in CRC and normal colorectal mucosa tissue and their possible relationship with the clinicopathological features of CRC,and analyze the relevance between KLF6 and WWOX protein, and then explore the roles of them in the development and invasion of CRC and their clinical significance.Methods1 The expression of KLF6 and WWOX protein was respectively detected by streptavidin-perosidase(SP) method in forty cases of CRC and forty cases of normal colorectal mucosa tissues, to analyze the relationship between gene expression and sex, age, differention, invasion, metastasis, and the relevance of these two gene expressions.2 Statistical analysis All the datas were analyzed by SPSS16.0 statistical package; The comparision of positive rate used the Chi-square test; The correlation between two variables used Spearman correlation analysis. The level of significant difference isĪ±=0.05.Results1. Immunohistochemistry results showed that, the positive rate of KLF6 protein in forty cases of CRC tissues was 45.0%(18/40), but in forty cases normal colorectal mucosa tissues was 82.5% (33/40), statistically significant difference (P< 0.05).2. The relationship between KLF6 expression and the clinicopathological of CRC. The expression of KLF6 had statistically significant difference in tumor differentiation, lymph node metastasis and tumor invasion (P< 0.05), however, there was no significant association in sex and age(P> 0.05).3. Immunohistochemistry results showed that, the positive rate of WWOX protein in forty cases of CRC tissues was 37.5%(15/40), but in forty cases normal colorectal mucosa tissues was 90.0%(36/40), statistically significant difference (P < 0.05).4. The relationship between WWOX expression and the clinicopathological of CRC. The expression of WWOX had statistically significant difference in tumor differentiation, lymph node metastasis and tumor invasion (P<0.05), however, there was no significant association in sex and age(P>0.05).5. Statistical analysis showed that the expression of KLF6 and WWOX protein in CRC was positive correlation (r=0.320, P<0.05).Conclusions1. The expression of KLF6 in CRC is low; And the expression of KLF6 is related to tumor differentiation, tumor invasion and lymph node metastasis,which indicate that low expression of KLF6 protein may be involved in the development and metastasis of CRC.2. The expression of WWOX in CRC is low; And the expression of WWOX has relationship with tumor differentiation, lymph node metastasis and tumor invasion, which indicate that low expression of WWOX protein may play an important role in the development and metastasis of CRC,3. The expression of KLF6 and WWOX is positively correlated, which indicate that WWOX may upregulate the expression of KLF6 by some mechanism; And low expression of them may jointly promote the development,invasion and metastasis of CRC; Combined detection of KLF6 and WWOX protein expression may be helpful to the diagnosis and evaluation of the degree of malignancy of CRC.
