The Study Anti-oncogene WWOX Expression In Patients In Colorectal Cancer

Objective To study the association between WWOX mRNA gene and the protein expression levels change in colorectal cancer , evaluate the clinico-pathological significance,and explore the effect of WWOX express on the development and progression in colorectal adenoma and colorectal tumor.Method: All cases are from the patients who have received surgery in Fujian Provincial Hospital. The colorectal cancer is sporadic excluding Hereditary Non-Polyposis Colorectal Cancer(HNPCC) and Familial Adenomatous Polyposis(FAP). The colorectal adenoma specimens from digestive endoscopy center of Fujian Provincial Hospital after endoscopic polypectomy。Tissue sample acquitted from patients were divided into three groups, colorectal adenocarcinoma group(n=45),adenoma group(n=45) and healthy person's mucosa group(n=20) ,Which come from surgical specimens with pathologically confirmed normal tissue (n=20) . Semi-quantitative RT-PCR was used to detect WWOX mRNA expression level changes;Immunohistochemical (EliVision? plus )detection methods for WWOX protein levels. Finally, the above results were tested byχ2and LSD-t(least significant difference) to analysis WWOX mRNA and protein different expression level in different tissues.Result:1. The expression levels of the WWOX mRNA(0.55±0.11)in colorectal adenocarcinoma patients was detected by semi-quantitative RT-PCR method, its expression levels were lower than those in colorectal adenoma(0.73±0.16)and normal tissue(1.04±0.13)(P<0.05).2.Immunohistochemistry showed that WWOX protein located in the cell cytoplasm, its positive rates of WWOX protein expression were 95.00%(19/20,65.89%(31/45),46.67%(21/45)(P<0.05)in normal tissue , colorectal adenoma and colorectal tumor respectively(P<0.05), with deep invasion and lymph node metastasis, the positive rate also will be dropped.Conclusion:Down-regulation of WWOX expression may be associated with the biological behavior and prognosis of colorectal cancer,and which may be used as an importantbiological marker and one of the signs of disease assessment.
