ObjectiveTo investigate the expression and significance of WWOX,p53 and Bcl-2 in nasopharyngeal carcinoma. Methods61 cases of nasopharyngeal carcinoma and 20 cases of chronic inflammation of nasopharyngeal mucosa patients with complete clinical and pathiological data,who were initially treated in our hospital during june 2009 to october.WWOX,p53 and Bcl-2 mRNA were evaluated by reverse trnascription polymerase chain reaction (RT-PCR),and its protein were measured by immunohistochemistry.The expression of WWOX,p53 and Bcl-2 in nasopharyngeal carcinoma and its clinical significance were studied.Results1.The expression of WWOXThe average expression of WWOX mRNA in nasopharyngeal carcinoma and chronic inflammation of nasopharyngeal mucosa are 0.69±0.34 and 0.89±0.49,the positive rate of WWOX protein in nasopharyngeal carcinoma (39.34%) was lower than chronic inflammation of nasopharyngeal mucosa(70.00%) (p<0.05). The level of expression of WWOX in stage III-IV nasopharyngeal carcinoma was lower than in stage I-II (p<0.05).The positive rate of WWOX was observed more lower in patients with neck lymph node metastasis (p<0.05). The positive rate of WWOX protein in nasopharyngeal carcinoma had no relation with the different sex,age and pathological classification (p>0.05).2.The expression of p53The average expression of p53 mRNA in nasopharyngeal carcinoma and chronic inflammation of nasopharyngeal mucosa are 0.88±0.59and 0.51±0.21, the positive rate of p53 protein in nasopharyngeal carcinoma (80.33%) was higher than chronic inflammation of nasopharyngeal mucosa(40.00%) (p< 0.05);The expression of p53 in stage III-IV nasopharyngeal carcinoma was higher than in stage I-II (p<0.05). The expression of p53 in nasopharyngeal carcinoma had no relation with the neck lymph node metastasis, different sex, age and pathological classification (p>0.05)3.The expression of Bcl-2The average expression of Bcl-2 mRNA in nasopharyngeal carcinoma and chronic inflammation of nasopharyngeal mucosa are 0.44±0.32 and 0.24±0.11, the positive rate of Bcl-2 protein in nasopharyngeal carcinoma (85.25%) was higher than chronic inflammation of nasopharyngeal mucosa(35.00%) (p< 0.05).The expression of Bcl-2 in nasopharyngeal carcinoma had no relation with the neck lymph node metastasis,different TNM stage,sex, age and pathological classification (p>0.05)4.Correlation between WWOX and p53,WWOX and Bcl-2In NPC,the expression of WWOX was negative correlated to the p53 and Bcl-2,respectivelly (p<0.05) Conclusion1. WWOX was lowly expressed in nasopharyngeal carcinoma,the expression was associated with TNM stages and neck lymphnode metastasis,suggest that the abnormal expression of WWOX maybe relevant with the process of happening, development and prognosis in the nasopharyngeal carcinoma.2. p53 was overexpressed in nasopharyngeal carcinoma,the expression was associated with TNM stages,suggest that the overexpressed of p53 maybe relevant with the process of happening and development in the nasopharyngeal carcinoma.3. Bcl-2 was overexpressed in nasopharyngeal carcinoma,the expression had no associated with TNM stage, neck lymph node metastasis, sex, age and pathological classification, suggest that the abnormality of Bcl-2 was an early event in the developpment and progression of nasopharyngeal carcinoma, may have little to do with metastasis and prognosis of nasopharyngeal carcinoma.4. The expression of WWOX was negative correlated to the p53 and Bcl-2 in NPC, suggest that in the development process of nasopharyngeal carcinoma, WWOX,p53 and Bcl-2 maybe interacted with each otherjoint or collaborative working.
