ObjectiveIn recent years,colorectal cancer become one of the most common tumour all over the world, It has become the major disease that threaten human health, but also one of the most common gastrointestinal cancer,abnormal cell proliferation and apoptosis may lead th the occurrence of colorectal cancer and progress. Survivin is recently identified members of inhibitor of apoptosis proteins (IAPS) which has unique structure,with the function of blocking the transduction of apoptosis signal by buliding to downstream molecules and inhibit its activity. KLF6 expresses on chromosome 10p,is a ubiquitously expressed zinc finger transcription factor that is part of growing family of Kruppel-like factors(KLF). The KLF family is broadly involved in differentiation and development, growth-related signal transduction, cell proliferation, apoptosis, and angiogenesis. The expression of KLF6 protein may be related with cancer development, and reduced expression in many cancer development.This research was designed to detection the expression of survivin and KLF6 protein in colorectal cancer, and analyze their correlation with clinical and pathological features and their relationship.MethodsThe expression of KLF6 and Survivin proteins were detected by immunohistochemistry in 68 colorectal carcinoma specimens and 40 normal colorectal mucosa specimens. The patients'clinicopathological parameters were collected and anlysised the relationship between Survivin and KLF6 protein in colorectal cancer pathological factors. The data were analyzed by SPSS 13.0 for windows. Resultsl.The positive expression rate of Survivin was significantly higher in colon cancer tissue(67.6%,46/68)than in normal colon tissue(2.5%,1/40) (P<0.01).2.Expressions of Survivin were not related to age, gender, histological type and histological differentiation (P<0.05) but related to depth of bowel wall invasion, lymph metastasis and Dukes staging.(P>0.05).3. The positive expression rate of KLF6 was significantly higher in normal colon tissue (44.1%,30/68)than in (82.5%,33/40) colon cancer tissue (P<0.05)4.The expression of KLF6 was found to be associated with differentiation, lymph node metastasis,tumor invasion and Dukes staging in colorectal carcinoma (P<0.05).5.A negative correlation was also noted between the expression of KLF6 and that of Survivin in colorectal carcinoma (r=-0.462, P<0.01).Conclusion1.The Survivin protein is over expressed in colorectal carcinoma than that in normal colorectal mucosa tissues;And the express of Survivin protein is related to depth of bowel wall invasion, lymph metastasis and Dukes staging,indicating that over expression of Survivin protein may be involved in the development and metastasis of colorectal carcinoma.2.The KLF6 protein is lower expressed in colorectal carcinoma than that in normal colorectal mucosa tissues;And the expression of KLF6 protein is related to differentiation, lymph node metastasis,tumor invasion and Dukes staging,indicating that low expression of KLF6 protein may be involoved in the development and metastasis of colorectal carcinoma.3.The expression of Survivin and KLF6 protein is negatively correlated, the over expressed of Survivin protein and low expressed of KLF6 protein may jointly promote the development,invasion and metastasisi of colorectal carcinoma;Combined detection of Survivin and KLF6 protein expression may be helpful to the diagnosis and evaluation of the degree of malignancy of colorectal carcinoma.
