Tremella fuciformis Berk is one kind of fungus and is famous for its' great nutritivevalue in our country. Whereas,Tremella fuciformis could not degraduate cellulose tonutrient by itself. Its' cohabitant fungus is nessisary in the process of fruiting bodygrowing development for the reason that the cohabitant fungus can turn woodflourmedium into all kinds of nutrient in the help of cellulase and some otherenzymes.This phenomenon has aroused rearchers'interest on it.However thetaxonomic position of cohabitant fungus has not been confirmed yet,and there is not aconclution about the relationship between Tremella fuciformis Berk and conhabitantfungus.This paper aims at studying a cohabitant fungus (S)of a wild Tremella fuciformisBerk(R) to ensure the cohabitant fungus's taxonomic position. Scanning electronmicroscope was used to view some microstructures.We use molecular biologicalskills to abtain the ITS sequence of879bp.We found that the band has99%similaritycompared with hypoxylon in NCBI Web.We abtain24ascospores from the wild cohabitant fungus (S).The mycelium growingspeed,colour changed time in PDA medium,mycelium shape,RAPD molecularmark and enzyme activity were studied inorder to analyse the polymorphism of the24ascospores.The results show that the mycelium growing speed of24ascospores differ both inPDA medium and woodflour medium,and the mycelium of all ascospores growsfaster in woodflour medium than in PDA medium. In the woodflour medium thefastest growing speed is0.73cm/d and the slowest is0.51cm/d whereas in the PDA medium is0.52cm/d and0.40cm/d. It indicates that different mediums have a cleareffect on the mycelium growing.We classify the24ascospores to5kinds by RAPDmolecular markers.After measuring the enzyme activity,we found all the ascosporeshave not the activity of amylase,endoglucanase and ligin peroxidase.And thedifference of both exoglucanase and β–Glucosidase among24ascospores is verysmall.By contrast,the laccase activity vary in24ascospores obviously.The greast gapreaches28enzyme activity units.At last,we cultivate Tremella fuciformis Berk(R) with24ascospores separately.All the Tremella fuciformis Berk(R) with24ascospores separately grow well and cansend up fruitingbody.This result indicated that the cohabitant fungus(S) is right thefungus that related with the development and output of Tremella fuciformis Berk'sfruitingbody.
