we are rich in Tremella fuciformis Berk,every year we produce the most Tremella fuciformis Berk in the world,Developing Tremella fuciformis Berk products of biological activity can strengthen the deep development of Tremella fuciformis Berk resources in our country,and make it more valuable.what's more,Tremella fuciformis Berk will be more useful,and developing Tremella fuciformis Berk products of biological activity can really do good to the comprehensive development and exploitation of Tremella fuciformis Berk and the Industrialization development of it.In this dissertation,the author conducted a series of study on Tremella fuciformis Berk which were produced through alkali extraction and acid hydrolysis.Samples with physiological effect are chosen,and their safety are confirmed by stimulative experiments.The results are as follows:1.A four-factors,three-level orthogonal experiment are done to find the best condition of alkali extraction,studying the factors which have effect on the extraction rate of polysaccharides such as material and extraction solution,pH,extraction temperature,and extraction time.The best condition are as follow:the temperature is 9℃,the pH is 9,material and the extraction solution is 1:100,and the time is 4 hours,under this condition,The yield rate of Tremella fuciformis Berk is 27.4%.2.A three-factors,three-level orthogonal experiment are done to find the best condition of acid hydrolysis,studying the factors which have effect on the hydrolysis rate of reducing sugar such as extraction temperature,extraction time and the concentration of hydrochloride.The best condition are as follow:The temperature is 100℃,the time is 3 hours,and the acid concentration is 5mol/L.Under this condition,The yield rate of Tremella fuciformis Berk reducing sugar is 73.87%.The Tremella fuciformis Berk polysaceharide extract is named YE,and the Tremella fuciformis Berk polysaccharide hydrolyzates are named YE1,YE2,YE3,YE4,YE5, YE6,YE7,YE8,YE9 by the yield rate of Tremellafuciformis Berk reducing sugar.3.The internal and external experiment for moisturizing effect shows that YE3 and YE5 both have excellent moisturizing effect.When Increasing the moisture content of the skin,they can also lower the volume of water lost from skin.4.YE9's inhibition rate on tyrosinase are 65.88%,and the inhibition rate on arbutin is 99.78%.YE9 have some whitening effect.The experiment on melanoma cells in mice shows that YE9's IC50 value are bigger than arbutin,and its effect on B16 cell survival rate are smaller than arbutin.YE9 is a safe inhibitor to melanoma.5.The examination of antioxidant effect shows that Tremella fuciformis Berk polysaccharide and Tremella fuciformis Berk polysaccharide hydrolyzates have some scavenging capacity to[DPPH-],The YE4 sample's scavenging rate to superoxide anion is 16.12%,but YE and other samples of Tremella fuciformis Berk polysaccharide hydrolyzates don't have scavenging capacity to superoxide anion. Besides,Tremella fuciformis Berk polysaccharide and Tremella fuciformis Berk polysaccharide hydrolyzates don't have scavenging capacity to hydroxyl radical.6.A test on Tremella fuciformis Berk polysaccharide and Tremella fuciformis Berk polysaccharide hydrolyzates by electronic microscope shows that YE have some hair protection efficacy.7.The experiment of inhibitory nonenzymatic glycosylation shows that YE, YE1,YE2,and YE3 have some inhibition effect on inhibitory nonenzymatic glycosylation,other samples of Tremella fuciformis Berk polysaccharide hydrolysis objects almost don't have inhibition effect on inhibitory nonenzymatic glycosylation8.Tremella fuciformis Berk polysaccharide and Tremella fuciformis Berk polysaccharide hydrolyzates have more effect on proliferation of Bifidobacterium than glucose.and YE5 has the best proliferation effect on Bifidobacterium..9.Choose sample YE which has hair protection efficacy,sample YE3 which has moisture efficacy,sample YE5 which has the efficacy to promote the proliferation of Bifidobacterium,sample YE9 which has the efficacy to inhibite tyrosinase.A stimulative experiment on them show that their hemolytic rate are all lower than 5%, and they have no hurt on membrane.So,they don't have irritant and can be used safely in both cosmetics and food.
