The axillaries buds of three breeds (P.suffruticosa'Wulongpengsheng', P.suffr -uticosa'Fengdanbai', P.suffruticosa'Taipinghong') were tested as explants. The effects of exogenous hormone on root induction were studied; the contents of endogenous hormones, IAA, ZR, ABA and activities of POD, PPO, and IAAO during the root development were determined; added different contents of ascorbic acid (Vc) to regulate the activities of PPO; regulated the environment such as light, darkness and low temperature to search the proper environment for rooting; studies on anatomical and morphological of roots were investigated why root-induction is difficult by paraffin sections, the results are as follows:1. The effects of exogenous hormone on root induction: the treatments of 1000 mg·L-1 IBA+1s and 300mg·L-1 IBA+10min are most proper for rooting and the average roots are high. The root rate of the treatment of 1000 mg·L-1 IBA+1s is 80% using P.suffruticosa'Tai pinghong', the root rate of the treatment of 1000 mg·L-1 IBA+1s is 35% using P.suffruticosa'Wulongpengsheng'. The rooting time of P.suffruticosa'Taipinghong'started form 4 to 6 weeks after rooting. The rooting time of P.suffruticosa'Wulongpengsheng'started form 5 to 7 weeks after rooting.2. The activities of endogenous hormones and enzymes during rooting.From the third day, the root primordium initiated, the activities of POD decreased the activities of PPO and IAAO increased. When the root protrusion out of the cortex of the shoot, the trends of the activities of POD became gentleness, the activities of PPO increased, the activities of IAAO decreased.From the third day, the root primordium initiated, the contents of IAA increased the contents of ABA and ZR decreased. Before the root protrusion out of the cortex of the shoot, the contents of IAA changed in volatile, the contents of ABA increased. When the root grow, the trends of the contents of IAA became gentleness, in the pre-grown period of the root, the contents of ABA decreased, in the late growth period of the root, the trends of the contents of ABA became gentleness, when the root growth ended, the contents of ABA increased greatly. When the root protrusion out of the cortex of the shoot and grow, the contents of ZR increased, in the late growth period of the root the contents of ZR decreased, when the root growth ended, the contents of ZR increased again.3. Added different contents of ascorbic acid (Vc) to regulate the activities of PPO: when the contents of ascorbic acid (Vc) increase, the activities of PPO decrease. The proper contents of ascorbic acid (Vc) of P.suffruticosa'Fengdanbai'and P.suffruticosa'Wulongpengsheng'is 50 to 100 mg·L-1 and 100 to 300 mg·L-1 separately. And the rooting situation is well.4. Regulate the environment of the rooting: when treated at 10℃for 10 days the P.suffruticosa'Wulongpengsheng'has high rooting rate 33.3% and the average roots is 1.8. The rooting rate of the treatment CK is 26.7%, the rooting rate of treatment 15 days and 20 day is 20%. The long time of low temperature will inhibit rooting. The rooting rates of the darkness treatment 9 days and 12 days of P.suffruticosa'Wulongpengsheng'are the highest, can reach 26.7% and the average root of treatment 9 days is the highest 3.5. So the darkness treatment 9 days is the most proper treatment. At the 4℃and darkness, the rooting rate of P.suffruticosa'Taipinghong'increases obviously. The treatment 3 days has the highest rooting rate and the rate is 66.7%. The longer the treatment is, the lower rooting rate is. The rooting rate of the red light treatment 10 days and 15 days of P.suffruticosa'Taipinghong'is 60%, the average root of treatment 15 days is 4, and the rooting situation is better than others. The rooting rate of the red light treatment 5 days and 10 days of P.suffruticosa'Wulongpengsheng'is 33.33%, the average root of treatment 10 days is 2.4. The rooting situation of P.suffruticosa'Taipinghong'is better than P.suffruticosa'Wulongpengsheng'because there are some differences between different varieties.5. Cytology observation on rooting culture of test-tube plantlet: the roots from the callus desquamate easier and the survival rate of transplant is lower because the conducting tissue did not connect with each other. The roots from the vascular bundle have some splits. Although their conducting tissues connect with each other, the survival rate of transplant is lower.6. The transplant of test-tube plantlet: using peat and vermiculite(3︰1),the survival rate is 32.4%. Using perlite and vermiculite(3︰1), the survival rate is 10%. The survival rate of transplant is lower, the color of the leavies which close to the seedlings survive easily. The longer the time of the rooting is, the higher the survival rate of transplant is.
