Using the the Neofinetia richardsiana Christenson seeds as explants, we select the suitable medium for germination of seeds, proliferation and differentiation of PLB,also select the suitable medium for rooting of the plant in vitro. Then compared the different methods of acclimatization, disinfection time of matrix and matrix ratio on survival rate of plantlets, and determinate all kinds of kinetic parameters changes of suspension cultures of PLB. Observed the morphology of seed germination process. The results are as follows.The optimal medium for the aseptic germination is 1/2 B5+6-BA 1.0 mg L-1+ NAA 0.2 mg L-1+AC 0.05%, the germination rate is 73.3%.The best liquid medium for suspension cultivation of PLB is MS+6-BA 1.5 mg L-1+NAA 0.5 mg L-1, the absolute growth rate is 260%; The best medium for differentiation of PLB is B5 +6-BA 1.0 mg L-1+NAA 0.2 mg L-1+0.15% AC, the differentiation rate is 87.5%. The study testified that the best rooting medium is MS+IBA 0.3 mg L-1+NAA 0.5 mg L-1, the rooting rate is 100%.Put the transplants acclimatization in the room for 2 days and then outdoor for 3 days, this hardening method to achieve the best effection,it can reach to 86%.The matrix which was transplanted with 1000 times carbendazim soaked for 5 min, the matrix is mixed with bark:moss:volcanic rock= 2:1:1 and the survival rate is as high as 90%.The process of increasing weight of PLB includes four stages:stagnant period for 0-6 d, growth stage for 6-12 d, stabilization period for 12-14 d and static phase for 14-16 d. During the process of cultivation, sucrose content decreases rapidly to 9.17 g-L-1 on the 8th day. On the contrary, the content of fructose rises gradually in the same time. We know that fructose consume fast at the last stage of cultivation, and total sugar present slow decline in the whole growth process. Phosphate decreases firstly before the 6th day, and then rise again; however, the content of phosphate declines slowly after the 10th day. Ammonium nitrate nitrogen declines sharp to 60.21 mg·L-1on the 6 day, and remains only 19.63 mg·L-1 on the 16th day. However, the changes of nitrate are relatively slow, since its content decreases slowly during the whole cultivation. The PH drop to 3.7 on the 8th day at first, while there is a slowly arise process at the late stage. the pH maintains 3.9 on the 6th day.After sowing the seeds of the first 10th day,part of the seeds start to swelling.On the 15th day, the spornioderm was filamentous off,and a large number of seeds began to germinate. On the 20th day,part of seeds began to change into PLB. On the 25th day,almost all the seed formation PLB.
