Abstract/Summary:
This paper reported the study of Camellia sinensis. Callus was inducted. Suspension culture of Camellia sinensis cell was carried out in 150 mL flask, then we constructed a non-structured kinetic model.Different explants,media, phytohormones, sucrose concentrations and pH could induce callus. The optimal conditions for the growth of Camellia sinensis callus was MS, combined with 1 mg/L 2,4-D, 1 mg/L NAA, 0.5 mg/L 6-BA and 3% sucrose. The pH of medium is 5.8. During 45 days cultures, the growth rate of callus was measured every three days. The curve of the growth shaped "S". Multiplication rate of callus was highest when calluses were cultured in the thirty-ninth day.In the course of suspension culture, the effects of rotation ratio, inoculum density, phytohormones, the bottle load and sucrose concentration were investigated. The optimal condition for the growth of Camellia sinensis cell was as follows: 100 rpm, 100 g/L FW, 3% sucrose; the reaction was carried out in 150 mL flask with the bottle load of 30 mL; the best medium was MS , combined with 2 mg/L 2,4-D and 0.25 mg/L KT.A non-structured kinetic model of the growth of Camellia sinensis cell suspension was constructed. This model described the rule of Camellia sinensis cell growth, limiting substance consumption and theanine production. These results showed that the model had a very high coincidence with the experimental dates.These equations of kinetic model were as follows respectively.
