Panax ginsing is the traditional Chinese medicine, which belongs to Acanthopanax gracilityusaceae Panax. It has some function such as invigoration, inhalation of air, promoting blood flow, to lengthen one’s life and so on. Ginsenosides are the active compounds. In recent years, how to increase the content of ginsenosides becomes more and more popular in modern Chinese medicine research.The experimental materials of this study obtained roots of Panax ginseng and Panax ginseng seed, to inducte, identifie and screene high yield ginseng hairy root suspension culture system by Agrobacterium rhizogenes A4.To establish a system in500ml flask of cell suspension culture for Panax ginseng, and then we constructed a non-structured kinetic model.The result showed:1. The optimal medium of ginseng hairy root for suspension culture:1/2MS.2. The optimum concentration of Carbon source、nitrogen source、phosphate:source is3%, NO3-/NH4+=2:1,PO43-is85mg/L.3. The optimal pH of ginseng hairy root for suspension culture is5.8, the growth、saponin content、and the matrix saponin were all maximum.4. During30days culture, the growth quantity of suspension hairy root was measured every three days. The curve of the growth shaped "S". Between27to30day biomass reached the maximum, the highest fresh weight is43.4g/flask, and the highest dry weight is2.32g/flask and saponin content is4.63%.5. In terms of culture, The optimal of table parameters:inoculum density is2.14g/L, the bottle load is256.88ml, rotation ratio is134.68rpm, culture temperature is22.36℃, dark culture.6. The consumption rule of medium was examined. Too low concentration of carbon source can not satisfied the need of suspension hairy root growth. And excessive concentration can inhibit hairy root growth.; The suspension hairy root use NH4+at first,and then use NO3-;The consumption of phosphate is relatively faster than other source.7. A non-structurd kinetic model of the growth of Panax ginseng hairy root suspension culture was constructed. This model described the rule of Panax ginseng hairy root growth, limiting substance consumption and saponin production.①Hairy root growth kinetic model:②Substrate depletion kinetic model:③Production formation kinetic model: The correlation coefficients R2of the three equations were0.99021,0.98908and0.98812, respectively, which indicated good correlation between experimental values and models. Therefore the equations could be used to describe Panax ginseng hairy root growth, saponin synthesis and substrate consumption process...
