Resveratrol and its glucoside isomers in wine have an important impact on its taste and quality. Developing a simple and accurate test method has been a hot research field in the analysis and evaluation of wine.A direct injection HPLC method was established for determination of four polyphenols containing wine. The effect of mobile phase composition, solvent strength, pH of the system and other factors on the separation of standards and wines were discussed. Common C18 column served as the stationary phase, the gradient was formed by acetonitrile and water and the instrument was a conventional HPLC. This simple and fast method was suitable for initial testing of general wines.A method for simultaneously determination of four resveratrol and its glucoside isomers in wine using SPE-HPLC-DAD was developed. Pre-treatment conditions on the samples results were explored. Combined with DAD spectra of secondary qualitative and multi-wavelength output, achieved the optimization of the chromatographic analysis. The recovery of targets ranged form 81.38% to 115.39%, relative standard deviation (n=7) ranged form 1.19%to 2.75%, and the detection limit ranged form 0.68 to 1.32 ug/L (S/N>3).The stability and antioxidant capacity of target compounds had been detected by the method of SPE-HPLC-DAD. The results indicated that trans-isomers unstable exposed to light and relatively stable at room trmperature; the total amount of resveratrol in the same type of wine can represent the quality of it. They are one of the fuctional components of wine.This developed SPE-HPLC-DAD method can effectively use the function of peak purity and multi-wavelength output with DAD detector, which could avoid the influence of impurities on the determination results. The different targets can be detected in the best wavelength. The data supported a certain reference value on the measurement and evaluation of wine.
