Resveratrol and its glucoside isomers in wine have an important effect on its qualityevaluation. Developing a simple and accurate test method has always been a hot researchin the analysis of wine.A HPLC method with direct injection was established for determination of resveratroland its glucoside in wine. The extraction of resveratrol and its glucoside in wine wascompared with organic solvent and aqueous two phase system.This paper has optimizedthe extraction conditions of different solid phase extraction columns which is used forextracting resveratrol and its glucoside in wine. This paper developed a method forextracting resveratrol and its glucoside isomers in wine by the solid phase extraction(SPE).The details were as follows:1. A HPLC method with direct injection was established for determination ofresveratrol and its glucoside in wine. The mobile phase ratio, pH of the mobile phase,detection wavelength, column temperature and other factors which affect separationefficiency of the standard substance of four kinds of polyphenols and wine sample wereinvestigated separately. The standard addition recoveries of resveratrol and its glucoside is93.69%-103.24%, the relative standard deviation is0.64%-3.24%, the detectableconcentration of T-PD and T-Res is0.3μg/L, the detectable concentration of C-PD andC-Res is0.7μg/L.The method has the advantages of convenient operation, and is suitablefor the detection of resveratrol and its glucoside.2. organic solvents were choosed in this study: Acetic ether: n-hexane(50:50/10:90/90:10),Aceticether:dichloromethane(50:50/10:90/90:10),Acetic ether,ethyl ether and chloroform. These organic solvents are used for extraction recovery ofresveratrol and its glucoside in wine. Compared the extraction ratio of resveratrol and itsglucoside in wine by ethyl acetate and ammonium sulfate aqueous ethanol aqueous twophase system. Ethyl acetate is better, it’s ratio of T-PD and T-Res can be over90%, but toC-Res, the ratio is relatively low.3.A SPE method to extract resveratrol and its glucoside isomers in wine wasestablished. Investigated the conditions of C18, PLS and PXA solid phase column, thesecolumns were used to extract resveratrol and glucoside in wine. The C18solid phasecolumn’s relative standard deviations were0.34%~2.32%and the recoveries were 78.54%~84.32%, the PLS solid phase column’s relative standard deviations were2.87%~4.69%and the recoveries were82.98%~90.06%, and the relative standarddeviations of PXA solid phase column were2.4%~3.5%and the recoveries were84.6%~108%.The precision and accuracy of this method is relative good.Different wine has different contents of resveratrol and glucoside. High quality winecontains lots of resveratrol and glucoside. But low quality wine has little or none at all.This study is to evaluate wine quality and distinguish the real wine from the fakes.
