This paper has established high performance liquid chromatography methods to measuretotal content of acetoin, cis-and trans-resveratrol content in wine. Also, a gas chromatographymethod for detecting the content of2,3-butanediol isomers in wine and distilled spirit wereestablished. The high-performance liquid chromatography was used to investigate backgroundcontents of total acetoin and cis-/trans-resveratrol in wine. The2,3-butanediol isomers contentof wine and distilled spirit were measured before and after irradiation by using gaschromatography method. These results above were used to identify the quality of wine and toidentify whether the wine and distilled spirit were irradiated or not. The main contents of thispaper are as follows:(1)The experimental conditions including pH value,2,4-dinitrophenylhydrazine (DNPH)content, reaction time, reaction temperature and processing step after derivatization, wereoptimize. Liquid chromatographic analysis method was established. The mobile phase wasdetermined as the composition of acetonitrile and water (55:45, V/V). Under the optimalconditions, in the range of0~40.0mg/L for acetoin was linear with correlation coefficient of0.9999. The spiked recoveries of acetoin in the dry red wine sample were in the range of95%~106%; the spiked recoveries of the dry white wine samples were in the range of94%~104%.The contents of total acetoin in different types of wine samples were tested.And the resultsshowed that the concentrations of total acetoin in domestic fake and shoddy wine samples weremuch higher than those in good quality wine samples. Besides,the concentration of total acetoinin home-brewed wine samples from bad grapes were also higher. Therefore, the concentration oftotal acetoin can provide a reference for the wine quality identification.(2)Recoveries of pretreatment methods were compared, which were direct injection, aceticether extraction, the solid-phase extraction, and acetic ether extraction combined withsolid-phase extraction. The direct injection method shows optimum recovery. The liquidchromatographic analysis method was established. The contents of cis-/trans-resveratrol inwines from different area, grape and year were tested. The relationships between them wereanalyzed. Also, the content of resveratrol in fake and shoddy wine samples and good qualitywine samples were compared. The obtained results showed that resveratrol content in wine can provide reference to the wine quality identification.(3)A chiral capillary column with gas chromatographic method was developed for thedetermination of three isomers of2,3-butanediol in liquor sample. The samples were extractedvia vibration with ethyl acetate and methanol(4:1, V/V) mixed solution. The extract was purifiedwith graphitization carbon black column using solid phase extraction device. The spikedrecoveries of three butanediol isomers were90%~113%in wine and96%~99%in spiritrespectively. The method was applied in the determinations of2,3-butanediol isomers in winesamples and sipirit samples before and after irradiated by Cobalt60-gamma rays with irradiationdose of10kGy. The results showed that d-2,3-butanediol was detected in spirit samples afterirradiation, but no obvious change of d-2,3-butanediol content occured in red wine, which couldprovide an evidence on whether the sipirit samples were irradiated or not.
