Serum Response Factor(SRF) is a highly conserved and ubiquitous transcription regular factor in mammalian, and its binding sites is a 10bp DNA sites ,called CArG-box, which have 1216 permutations. SRF was found in research to c-fos , and because of its connection with cancer and heart diseases and so on , it becomes a hot topic in modern time.A lot of paper has published which are focused on the function of it and its cofactor in metabolism, but few about its binding sites. This article is mainly about CArG-box in the scale of genome by the dataset of Chip-on-chip, as it supply four cell lines ,I can also do some work about the different regulation level in different cells.I use the dataset of GSE8489 which was part of ENCODE that was launched by NHGRI as cut sets, and GRCh37 from ENSEMBL as rough data. Then I use several method to search the potential motif, such as consensus , PSSM. Finally, I got such conclusion:First ,transcription regular factor plays different roles in different cell line.Second ,PSSM is the better way to search motifs.Third ,different environment in organisms will influence regulation seriously.Fourth, SRF will bound to A/T rich region if environment of biology allow.Fifth, different multi in different cell.
