Field Effect Transistor(FET)cell sensor is a new type of sensor which can monitor the electrical signals of in vitro cultured cells on real-time.Among all kinds of cell electrophysiological signal detection methods,it has the characteristics of simple operation,high-throughput,minimal interference and stimulation to cell,and sustained signal acquisition.Micro-nanofabrication technology involves a variety of heavy metals and corrosive chemical reagents.Considering the potential toxicity of its raw materials and manufacturing process,the products produced by this process must undergo toxicity testing to ensure its good biocompatibility.In this paper,a series of work has been done on the design of the sensor chip based on the physiological and biochemical characteristics of cells;In toxicity test,rat hippocampal neurons were selected to be cultured on the different thickness of SiO2 coated sensor chip,compared with the cells cultured on Corning standard 24-well plate.Qualitative observation of microscopy and quantitative test of Thiazolyl Blue Tetrazolium Bromide(MTT)were used to evaluate the toxicity.The main conclusions of this paper are as follow:1.A p-channel FET sensor is designed,which power supply voltage is-3?1V,the conduction current is 30-60 ?A,the leakage current is less than 1 nA,the channel resistance is 10-20 K?,the single effective sensing area is 2.5 ?m2.And it has good transfer characteristics.It can adjust the effective working range by changing the back gate voltage.2.A 10*10 sensor array chip is designed and fabricated,the secondary coding of the sensor array is completed,the software thermodynamic diagram is drawn,the corresponding relationship between the chip and the software is constructed,and the signal is displayed in the most intuitive way.3.The assembly of the first generation FET culture dishes has been completed.According to the characteristics of the chip and other devices,the process of cleaning with low frequency ultrasound for 5 minutes and then high frequency ultrasound for 10 minutes in 50? water has been designed.The process of cleaning and sterilizing can meet the practical needs after several successful cultures of hippocampal neurons.4.It is proved that the FET culture dishes coated with SiO2 insulating layer of 30 to 100 nm thickness were all grade 1 in MTT evaluation index and had no biological toxicity.With the increase of the thickness of SiO2 insulating layer,the survival rate of hippocampal neurons cultured shows an upward trend...
