Objective: To isolate the signal products of Maillard reaction from processed Cibotium baronetz(L.)T.sm.; Identified that whether the Maillard reaction happens logically in the procedure of Cibotium baronetz(L.)T.sm; Investigated the transformation of chemical constituents between raw and proceed product and the relationship of transformation of chemical constituents and pharmacodynamic active. On the basis of above-mentioned study, drafted the quality criteria of processed Cibotium baronetz(L.)T.sm, to ensure the validity and safe of clinical medicine.Material and method:Material: The dried rhizome of Cibotium baronetz(L.)T.sm. processed Cibotium baronetz(L.)T.sm.Method:1. TLC and HPLC were carry out to analyze the variation of chemical ingredients between raw and processed Cibotium baronetz(L.)T.sm. and study the impact factors of procedure to chemical constituents.2. The chemical constituents changing significantly after processing were isolated and purified by various chromatographic methods including silica gel column chromatography, Sephadex LH-20 column chromatography and HPLC. Their structures were identified by the physico-chemical property and spectroscopic data.3. The chemical ingredients changing remarkably in Cibotium baronetz(L.)T.sm. after processing were identified by HPLC.4. On the analysis of reagents, condition, products and the property of products, the Maillard reaction which happened during processing was reinforced logically.5. The relationship of transformation of chemical constituents and pharmacodynamic of proceed Cibotium baronetz(L.)T.sm. was studied by means of Osteoporosis.6. The quality criteria of proceed Cibotium baronetz(L.)T.sm. was established by HPLC and TLC.Result:1. The variation of chemical constituents before and after processing showed that the chemical constituents in the chloroform, ethyl acetate and n-butanol extract of crude and processed drug all changed remarkably, especially the phenols in the extract of n-butanol.2. Ten compounds relative to Maillard reaction were isolated from the extract of n-butanol and identified as species of furfurol,γ-pyrone and phenolic acid.3. The chemical constituents changing remarkably after procedure were that 2(Hydroxy acetyl)faran, maltol, 5-hydroxy-maltol, Hydroxymethyfurfural, [5-formacyl furfuryl] ether ,Protocatechuic aldehyde, Kojic acid, Protocatechuic acid, 3(3,4-Dihydroxy phenyl)-2-propenoic acid,α-D-Glucose.4. The fact that the Maillard reaction happened during processing was confirmed.5. The pharmacodynamic study showed good correlation between the chemical constituent and activity.6. The content of protocatechuic acid in processed Cibotium baronetz(L.)T.sm. was determined by HPLC and the quality criteria of Cibotium baronetz(L.)T.sm. was established.Conclusion:1. The chemical constituents changing remarkly before and after processing of Cibotium baronetz(L.)T.sm. existed definitely.2. The extractant of proceed Cibotium baronetz(L.)T.sm. contained the chemical ingredients belonging to species of furfurol and pyrone.3. Maillard reaction happened during the procedure of Cibotium baronetz(L.)T.sm..4. A good correction between chemical constituents and activity was confirmed.
