There exists only one species of Cibotium barometz(L.)J.Sm. in the China mainland. Its'leaf flavonoid compounds have rarely been documented by now. The current study aims to develop a procedure for extraction and purification of total flavonoids from the leaves of Cibotium barometz and to investigate its'antioxidant properties. The main results include:(1)Flavonoids from the leaves of Cibotium barometz was extracted by using the microwave-assisted bath extraction technique.The research concerns on the extraction efficiency which was relate to the concentration of alcohol,the ratio of material to liquid,irradiation time,microwave power,bath temperature and water bath time.According to the result of single factor tests,the concentration of alcohol,irradiation time,microwave power and bath temperature were chose as the factor to examine.The orthogonal experiment use L9(34) orthogonal table and the best conditions for the extraction were as follow:40% ethanol,microwave power of 160W,irradiation time for lmin,the ratio of material to liquid of 1:45,water bath time for 0.5h,bath temperature maintains at 90℃,the rate of total flavonoids extraction was 11.96%.(2)The characteristics and technology of four types of macroporous resins were compared to separate total flavonoids extracted from the leaves of Cibotium baromnetz.The results showed that DA-201 resin,with good absorbability and desorption properties,was the best one for purifying the total flavonoids in the leaves of Cibotium barometz.The following were the best technical parameter of its static adsorption:the ratio of resins and extracts was 1:5 (W/V),pH value was 3,the time of shaking at constant temperature was 2h and the concentration of eluant was 60% alcohol.In dynamic desorption,the best technical parameter was expressed as follow:the eluant flow rate was both 1.0mL/min and the volume of the eluant was 3 times of the resin bed.(3)Favonoids extracted from the leaves of Cibotium barometz deoxidize activity test showed the effect of deoxidize activity was intensified while the addition of flavonoids increased,(flavonoids addition between 0.04-0.20mg).when flavonoids addition was 0.4mg, the effect of scavenging hydroxyl free radical and superoxide anion free radical were citric>flavonoids, The effect of scavenging the two free radical was intensified while the addition of flavonoids increased,(flavonoids addition between 0.4~2.0mg).
