Respect Kidney Zhuanggu Soup Contributed To The Study Of Bone Formation Around The Prosthesis

Objective:To observe the content of the tumor necrosis factor-α(TNF-α) and the morphological changes of bone tissue about the effect of the zun-shen-Zhuang-gu Tang to rabbit that inpiant kirschner into femoral.Methods:24 healthy adult rabbits (male and female are not limited) , divided into two groups randomly:"A"group for the experimental group, and"B"group was the antithesis group,12 in each group. The two groups are implant Kirschner which diameter is 4mm in the right knee, model after successful, A group fed with Zun-Shen-Zhuang- Gu Tang , 20ml each time, twice a day; the B group fed with saline; 20ml each time, twice a day;per gavage 7 days suspended two days. the two groups are injection of polyethylene particles 1.5ml at 2,4,6,8 and 10 weekend in the right knee joint.12 weeks later,animals were killed,check femoral metaphyseal and shaft bone tissue (size is about 1.5cm * 1.5cm * 0.5cm). The metaphyseal bone tissue parcel with foil and frozen in liquid 24 hours, triturate as far as possible and Centrifugal,use supernatant to check TNF-α; metaphyseal and the shaft bone are soke with alcohol that Concentration is 40% immediatly,and then dehydrate in 40%, 70%, 80%, 95% and 100% alcohol,transparent in xylene, methylmethacrylate -embedded fixed. Heavy slicer slice, analysis with Leica FW4000 analysis software.Results:Each group have one rabbit daid. 6 specimens, around the kirschner, are layer of fibrous tissue in the B group, while there is only one specimens in the A group, the others bone tissue prosthesis close to or have a layer of new bone tissue around; The B group, there are two kirschner enter into bone marrow cavity,but A group, all kirschner are outside of bone marrow cavity;The content of TNF-αabout two groups is significant different;Bone tissue morphology indicators:the wide osteoid,osteoid surface, the average distance tetracycline double-labeled line and bone mineralization appositionrate are significant difference between two groups.Conclusion: The content of TNF-αabout two groups is significant different;At the point of morphological changes view, the role of promote osteoblast about Zun-shen-Zhuang-gu Tang is obvious, worthy to be popularized.
