Objective:This study aimed to investigate the effect of Astragalus extract on inhibiting rate of hydroxyl radical(·OH)and activity of glutathione peroxidase(GSH-PX) in skin of natural aging mice and to observe the ultrastructure of natural aging mice skin and the perfection of Astragalus extract in ultrastructure.Thus verifying and exploring the role of Astragalus extract in delaying natural aging from the following 2 respects:the clinical significance of anti-aging medicine in Chinese medicine and the free radical damage theory of Western medicine.Material and Method:12 4-month-old healthy female Kunming mice,as the young control group(A),60 18-month-old healthy female Kunming mice were randomly divided into 5 groups:the elder control group(B),low-dosage Astragalus group(C),medium-dosage Astragalus group(D),high-dosage Astragalus group(E),vitamin EC positive control group that was old age group (F),there were 12 mice in each group.Group A and group B were administered with saline(20ml/kg),group C,D and E were administered with Astragalus extract of 4g/kg,8g/kg,16g/kg respectively;group F was administered with vitamin EC Suspension of 3mg/kg.Gavaging once a day,the inhibition rate of·OH and the activity of GSH-PX in skin tissue were measured by biochemical method on the 31st day and the changes of organizational structure of mice skin were observed.Results:1.Compared with the young control group,the inhibitory rate of·OH in other 5 groups were significantly reduced(respectively is:P<0.01,P<0.01,P<0.01, P<0.01,P<0.01),the activity of GSH-PX were significantly low(respectively is:P<0.01,P<0.01,P<0.05,P<0.05,P<0.05),which indicates that the inhibition rate of·OH and activity of GSH-PX in skin of natural aging mouse were significantly reduced with age.2.Compared with the elderly control group,the positive elderly control group and elderly Astragalus group inhibiting rate of·OH enhanced(respectively were:P<0.05,P<0.05,P<0.01,P<0.01),the activity of GSH-PX increased (respectively were:P<0.05,P<0.01,P<0.05,P<0.05).which show that vitamin EC suspension and Astragahs extract can increased the inhibiting rate of·OH and activity of GSH-PX in the skin of mice,therefore Astragalus has a function of delaying skin senesconce.3.Compared elderly positive control group with elder treatment group:the inhibion rate of·OH in skin of mice in high-dosage of Astragalus extract group was superior to that in the positive control group(P<0.01);the datal shows that Astragahs extract can improve the inhibition rate of·OH of skin tissue, and were more effective than vitamin EC suspension group.In enhancing activity of GSH-PX of skin tissue,the medium-dosage of Astragalus extract group were significantly better than the positive control group(P<0.05),which indicates that Astragahs extract can improve the antioxidant capacity of aging mice,the effect of medium-dosage of Astragalus extract is superior to the vitamin EC suspension.There are no significant difference among different dosage of Astragalus extract groups,P>0.05.4.According to histomorphological observation:typical Langerhans granules can be seen in the young control group,disappeared in the elderly control group and a few in high-dosage Astragahs extract group.Conclusion:1.Astragalus extract can enhance the inhibiting rate of hydroxyl radical(·OH) and the activity of ghtathione peroxidase(GSH-PX) in skin of natural aging mice.2.According to morphological observation:compared with the young control group, skin of natural aging mice were senecence whit age and Astragahs extract can effectively slow down the aging state of the skin.3.Enhance the righteousness is an effective way to anti-senecence of skin.
