The trditional Chinese medicine especially the astragalus has the obvious ability in eliminating and delaying exercise induced fatigue. But there is not much in experimental study on the respects of delaying fatigue, improving the ability of exercise body and influencing the free radicals, and the exact mechanism is still not perfect. The experiment analyses every groups of the related biochemical index in rat serum by intragatric administration with astragulas extracts and positive control drug (Lycopene), in the same time, the erperimental rats were needed to have acute endurance inelastic intensive exercise with moderate degree for the sake of researching the impact of astragalus on rat oxygen free radical metabolism, and it will have a preliminary dicusses on its mechanism in this experiment.Objective:A research on the effect of astragalus root extract and move-ments on free radical metabolism in rat serum.Research Methodology:Fifty Rats were divided at random into five groups (n=10 in each group):astragalus low-dose group B1, astragalus middle-dose group B2, astragalus high-dose group B3, positive drug control groups C and blank normal group A. Astragalus low-dose, middle-dose and high-dose groups were supplemented with the same dose of astragalus root extract for 21days(Bl:2.5g/kg/1dB2:5g/kg/1d B3:10g/kg/1d), positive drug control group was supplemented with lycopene for 21days(C:5mg/ld), blank normal group was free to drink water every day, and all groups were free to eat during the period of taking medicine. And then, all rats would have a disposable weight loading swimming until they felt utterly exhausted after those 21 experimental days, and loaded weight of the rats occupied 4% of their body weight. All rats need to be taken blood instantly through their femoral after exhaustive exercise, the blood samples were put on the table 1 hour for blood coagulation first, and then centrifugated at 4000 rpm/min for 10 minutes in a centrifuge, measured the changes of the indicators shown about in each group:T-AOC, CAT, GSH, SOD AND MDA in serum.Result:1.Compare the astragalus group and positive drug control group to blank normal group, the time of swimming exhausition of rats had a significant improvement compared with the blank normal group, and the time of swimming exhausition of these groups (B1,B2,B3) increased regularly. The time of swimming exhausition of the astragalus and positive drug groups were obviously longer than the blank normal groups;2.In comparison with the blank normal group, it was found that the variation of SOD in serum increased significantly in the astragalus group and positive drug control group. By comparing with B1, B2 and B3, experimental results showed that the SOD content of serum in B2 was slightly higher than in group B1, and the SOD content of serum in B3 was obviously higher than B1 and B2;3.The content of GSH in serum was significantly increased in the astragalus group and positive drug control group by comparing the blank normal group. When it comes to the comparisons among these groups of C, B1, B2 and B3, the GSH content of serum in group C was significantly higher than that group B1 but lower than group B2 and B3. By comparing with B1, B2 and B3, experimental results showed that the GSH content of serum increased regularly, and statistical difference existed among group B1, group B2 and group B3;4.In comparison with the blank normal group, it was found that the variation of CAT in serum increased significantly in the astragalus group and positive drug control group. When it comes to the comparisons among these groups of C,B1, B2 and B3, no significant difference of the CAT content of serum was found between group C and B1, and the CAT content of serum in C was slightly lower than B2 but significantly lower than that of in group B3. By comparing with B1, B2 and B3, the results showed that the CAT content of serum decreased regularly but no obvious change among them;5.Compare the astragalus group and positive drug control group to blank normal group, the content of MDA in serum of rats had an obvious decreased compared with the blank normal group. By comparing with C, B1, B2 and B3, the MDA content of serum in C was slightly higher than B2 and B3 but lightly lower than that of in group B1. In comparison with the MDA content of group B1, B2 and B3, no significant difference was found between group B2 and group B3 but both of them were lower than group B1;6.The content of T-AOC was significantly increased in the astragalus group and positive drug control group by comparing the blank normal group. When it comes to the comparisons among these groups of C, B1, B2 and B3, the GSH content of serum in group C1 was lower than all those groups B1, B2 and B3 but no significant difference among them. By comparing with B1, B2 and B3, no significant difference of the T-AOC content was found between group B1 and B2, and the results showed that the T-AOC content in group B3 was slightly higher than in group B1 and group B2 but no obvious change among those three groups.Conclusion:The antioxidation ability in rat was significantly increased after taking the astragalus root extract, the MDA level was significantly decreased in this experiment. This result indicated that the astragalus root extract not only can effectively raise antioxidant ability and the elimination ability of free radical, but also lipid peroxidation and oxidative stress were caused by motion would be relieved to some extent.
