Objective:According to the characters of Chinese medicine,this topic adopted the technology which contained several subjects to study the therapeutic material basis which is about curing diabetes in Leontopodium leontopodioides(Willd.)Beauv;then,we can establish the standard of quality control in medicinal substances.And the standard is relative with the curative effect closely.Material and Method:Basing on the relative reports,we analyzed chemical composition in Leontopodium leontopodioides(Willd.)Beauv.,through out the Physicochemical Identification;according to the results of the Physicochemical Identification,we utilized the the method of solvent polar extract to prepare the different polar extract which was come from Leontopodium leontopodioides(Willd.)Beauv.;we adopted the HPLC method and utilized the DAD detector, to establish chromatographic condition which contained the most amount of information in different polar compound;we used the knowledge about Pharmacodynamics to investigate effect between different polar compound and BG,HBA1C in rats which were leaded to the Diabetes models through out the high fat diet and streptozotocin;we correlate the Fingerprints of Leontopodium leontopodioides(Willd.)Beauv,with the Pharmacological activity,using the methods of Correlation Analysis,cluster analysis and Principal Component Analysis,to study the therapeutic material basis about Anti-diabetic in Leontopodium leontopodioides(Willd.) Beauv.And then we utilized methods of Microscopic identification,TLC and HPLC to establish quality evaluation mode which was based on the special effect in Leontopodium leontopodioides(Willd.) Beauv.Results:(1) Through out the experiment of Physical and chemical identification of the chemical constituents we knew that it contained some kinds composition,just like phenolic acid,polysaccharid,saponin,Alkaloids and flavonoids and so on.(2) Through out the investigation of detection wavelength,different kinds of chromatographic column and different mobile phase system,we identified the final chromatographic condition which contained the most amount of information in different polar compound.For example, Octadecyl bonded silica;0.2%Phosphoric acid aqueous solution-Acetonitrile gradient elution; Flow rate 1.0mL/min,Detection wavelength 290nm,Column temperature 25℃,And then, we used the method of HPLC to get 36 peaks from three different kinds of polar extracts.(3) Through out the experiment of effect between different polar compound and BG,HBA1C in rats which were leaded to the diabetes models through out the high fat diet and streptozotocin,its result showed that in the two indexes of BG and HBA1C,the water extract group and n-butanol extract group had the obvious effect on lowing the value of BG and HBA1C.(4) We correlate the Fingerprints of Leontopodium leontopodioides(Willd.)Beauv,with the Pharmacological activity,by using the methods of Correlation Analysis,cluster analysis and Principal Component Analysis,to know that there was a significant negative correlation between 14 peaks area and the values of BG and HBA1C.And there was a significant positive correlation between 10 peaks area and the index of inhibitory rate of BG.Conclusion:(1) Through out the experiment of Physical and chemical identification of the chemical constituents we knew that it contained some kinds composition,just like phenolic acid,polysaccharid,saponin,Alkaloids and flavonoids and so on.In them,Saponin and Alkaloids were never reported.The research of Saponin and Alkaloids need to wait the further experiment in the future.(2) Through out the experiment of effect between different polar compound and BG,HBA1C in rats which were leaded to the diabetes models through out the high fat diet and streptozotocin,its result showed that the water extract group and n-butanol extract group have the obvious effect of lowing the values of BG,HBA1C.At the same time it has the effect of inhibited hyperglycemia.In a while,we found that intraperitoneal injection of high fat diet combined streptozotocin-induced diabetic rat model has no obvious effect to the islet function. So the index of serum insulin is not fit for this model.(3) We correlate the Fingerprints of Leontopodium leontopodioides(Willd.)Beauv,with the Pharmacological activity,by using the methods of Correlation Analysis,Cluster analysis and Principal Component Analysis,to know that there was a significant negative correlation between 14 peaks area and the values of BG and HBA1C.And there was a significant positive correlation between 10 peaks area and the index of inhibitory rate of BG.So the 16 peaks(in them,there were 8 peaks to be contained in the above two categories of indexes)above were the therapeutic material basis.In them,X7 was protocatechuic acid;X11 was protocatechualdehyde.(4) By determining the content of protocatechualdehyde and protocatechuic acid,we can understand that there are different contents in Liaoning because of the different habit, collecting time and so on.But in the south,the contents have large difference,comparing with in the north.
