Xiaochengqi decoction was reported originally in shanghanlun, written by Zhang Zhongjing. In the clinical practice of traditional Chinese medical science, the famous decoction has been used to treat constipation over a very long period of time and has produced quite a good effect.The compatibility and therapeutic basis of Xiaochengqi decoction were studied in this paper. Xiaochengqi decoctions of different extracting methods were prepared. Defecation effects on mice was studied with above solutions. As a result, 70 % alcohol was selected as the optimal extraction reagent.The quantitative analysis of the compatibility was accomplished by design of combinations of Xiaochengqi decoction. Pharmacological experiments were performed according to 4 combinations of Xiaochengqi decoctions. The reasonability of the compatibility of Xiaochengqi decoction was elucidated in this paper.RP-HPLC method with UV detection was developed to obtain fingerprints of 4 combinations. Areas of 35 peaks were selected as chemical data. Correlation analysis was applied to interrelating the chemical and pharmacologic data. 4 chemical constituents in Xiaochengqi decoction were defined as therapeutic material basis of it. Among them, Rhein in Radix et Rhizoma Rhei , Magnolol in Cortex Magnoliae Officinalis and Hesperidin in Fructus Aurantii Immaturus were selected as the quality control indices of Xiaochengqi decoction.A method for determination of Rhein ,Magnolol and Hesperidin in Xiaochengqi decoction by HPLC with UV detection was established. The sample was separated on a Thermo-C18 (4.6 mm×250 mm i.d., 5μm) column using methanol-1% acetic acid solution (72:28, v/v) as the mobile phase at the flow rate of 1.0 mL·min-1.
