Circulation Line Water Antagonistic Renal Interstitial Fibrosis In Rats,

OBJECTIVE:To study the effects of rivalrying the progress of rat renal interstitial fibrosis by the Huayu Xingshui recipe.METHODS:The rats were randomly divided into unilateral ureteral ligation Testing Group and sham operation Control Group.After being ligated,the rats were further randomly divided into the model group, Huayu Xingshui recipe treatment group(Chinese herbal group) and Captopril treatment group.After respective medication by placebo, Huayu Xingshui herbals and Captopril for 7days,14 days,21days and 28 days,we executed the rats respectively,took out of the obstructive kidney, stained the renal tissues by Masson,immunohistochemistry and hybridization in situ methods,and applied image analysis system to observe the renal interstitium's pathological change degree semi-quantitatively,the expression of MMP-2 and FN mRNA in the renal interstitium.RESULTS:Compared with the Control Group,Collagen of the renal interstitium,the expression of MMP-2 and FN mRNA in the renal interstitium of the Testing Group were increased(P＜0.05 or P＜0.01). After 7 days medication,Collagen and expression of FN mRNA in the renal interstitium were more decreased in Huayu Xingshui recipe treatment group and Captopril treatment group than the model group(P＜0.05).But the expression of MMP-2 in the renal interstitium didn't show the difference of statistical significance between the testing groups.After 14 days,21 days and 28 days medication,Collagen and the expression of FN mRNA in renal interstitium were further significantly decreased in Huayu Xingshui recipe treatment group and Captopril treatment group(P＜0.01).But they didn't decline to a level of the Control Group(P＜0.01).The results also showed that the expression of MMP-2 in renal interstitium was increased in Huayu Xingshui recipe treatment group and Captopril treatment group compared with the model group(P＜0.01).CONCLUSION:Compared with the model group,Huayu Xingshui recipe treatment could decrease Collagen and the expression of FN mRNA in renal interstitium of unilateral ureteral ligation.It could also increase the expression of MMP-2 in the renal interstitium.The results showed Huayu Xingshui recipe can rivalry renal interstitial fibrosis.
