| Objective:To investigate the mechanism of Jing Kang Ling(JKL) capsule improving the ultrastructure of cervico-skeletal muscle of cervical spondylosis(arthro-myodynia type) rabbits model,and provide data for the practical use of JKL,by observing the effects of JKL on ultrastructure of cervico-skeletal muscle of cervical spondylosis (arthro-myodynia type)rabbits model.Methods:60 Japanese rabbits were randomly assigned to six groups:high dose JKL treatment group,medium dose JKL treatment group,low dose JKL treatment group, JingFuKang control group,model control group,and blank control group(10 rabbits in each group,with half of male and half female).Except the blank control group,50 rabbits model with wind-cold-dampness type cervical syndrome is set up by long-term head down position and wind-cold-dampness stimulation.Using Jing Fu Kang as positive control,JKL is given to rabbits model in different doses for 4 weeks.Three days after treatment,do the examinations: observe the Histo-pathological changes of histologic sections from skeletal muscle, which stained with hematoxylin and eosin(HE) and the ultrastructural changes of skeletal muscle by transmission electron microscope(TEM)。Results:1.Light microscopic observation:muscle fiber atrophied and degenerated,and the vascular cavity grows down in control group.The morphological changes of ultrastructure in high,medium,low dose JKL treatment groups and Jing Fu Kang treatment group are better than the control groups.The changes in high,medium dose JKL treatment groups and Jing Fu Kang treatment group is much better than low dose JKL group.2.Ultrastructure observation:In control group,muscle fiber atrophies,cell organs such as mitochondria,muscle rod degenerate,glycogen particles grow downwards.The morphological changes of ultrastructure of high,medium,low dose JKL treatment groups and Jing Fu Kangtion control group are better than the model control group. The changes in high,medium dose JKL treatment groups and Jing Fu Kang treatment group is much better than low dose JKL group.Conclusion:1.Long-term head down position and wind-cold-dampness stimulation damages the musculi colli tissue of the rabbit cumulatively,inducing a series of histopathological changes;2.JKL can make muscle ultrastructure recover from damages,impacts the mitochondria and muscle rod obviously and increases the muscle glycogen content.The mechanism is probably related with that JKL can improve the micro-circulation of muscular tissue. |
