Objectives: To study the content changes (the total alkaloids,ester diterpenoid alkaloids, diester diterpenoid Alkaloidas and the relevant constituents) after compatibizing White Prepared lateral Radix of Aconite and Dried Ginger in the different environments. At the same time, with the analysis of compatibility by TLCS and HPLC, we establish the chemical library of active ingredients and toxic ingredients , identify the material foundation and the basis properties of toxic medicine, explore the science and rationality of processing and compatibility.Methods: we evaluate the change of active ingredients and toxic ingredients in the compatibility of Radix Aconite Lateralis Preparate in the the different environments of decoction, artificial gastric juice and artificial intestinal fluid by Multiple Indicator system .Moreover,we use acid dyes colorimetric and improved hydroxamic acid iron spectrophotometry to determinate the contents of total aconite alkaloids and ester-alkaloid .we determinate the contents of alkaloids and phenols by HPLC , and determinate the change of contents related in the petroleum ether location, ethyl acetate location, n-butyl alcohol location after different proportion of Compatibility by TLCS and HPLC.Results: 1. Compatibility in medicine: After Compatibility, Different proportion of Dried Ginger and Radix Aconite Lateralis Preparate have effect on the contents of total alkaloids .The contents of total alkaloids is lower than that after hatching in artificial gastric juice and artificial intestinal fluid, Moreover, after Compatibility, the contents of ester-alkaloid is as higher as the numer of Radix Aconite Lateralis Preparate ,but the contents of ester-alkaloid is higher than that after hatching in artificial gastric juice and artificial intestinal fluid, Compatibility between different proportion of White Prepared lateral Radix of Aconite and Dried Ginger has greater effect on double-ester alkaloid content which is lower,and the stability of them is hypaconitine> new aconitine> aconitine .The contents of double-ester alkaloid is obviously lower after hatching in artificial gastric juice and artificial intestinal fluid ,but the stability of hypaconitine and new aconitine is improved,furthermore it is in arificial intestinal fluid obviously. Contrary to double-ester alkaloid ,the contents of phenols in Dried Ginger is as lower as the number of White Prepared lateral Radix.Compatibility in the effective parts: Compatibiliting extract from Dried Ginger and the total Aconite from alkaloid Radix Aconite Lateralis ,we found that the contents of total Aconite is as lower as the numbe of extract from Dried Ginger, so, we can drow conclusion that extract from Dried Ginger has obviously effect on the total Aconite ,which can inhibit the dissolution of total Aconite, and the the change trend of ester-alkaloid and double-ester alkaloid is the same as the total Aconite 2. After Compatibility in different proportions, in the TLCS,compared to Dried Ginger, the peak areas are biger or smaller as the number of White Prepared lateral Radix in the petroleum ether location, n-butyl alcohol location,in addition to some peaks partly increase or partly decrease in ethyl acetate location, there is a new peak, and the separation and capacity of the points good.After phosphomolybdic acid color,it's color is dark blue, and after vanillin significant color, it is yellow.The contents of new peak isn't increasing as the number of White Prepared lateral Radix ,so we infer that the new composition is a intrinsic composition in Dried Ginger,which can be show up afer interaction with White Prepared lateral Radix .3. After Compatibility in different proportions, in the HPLC,compared to Dried Ginger,a peak area is smaller as the the increase of the number of White Prepared lateral Radix in the petroleum ether location, in addition to two peaks partly increase or partly decrease in ethyl acetate location, there are two new peaks, contents of whicn don't increase as the number of White Prepared lateral Radix .otherwise ,there isn't big chane in the n-butyl alcohol location. The compositions of extraction before and after compating are nonlinear, particularly the emergence of the new peak,whicn can provide some proof to mutual promotion between Prepared lateral Radix of Aconite and Dried Ginger...
