Prepared Slices of Radix Scrophulariae is the round andelli-ptical flake of Scrophularia ningopoensis Hemsl’s dried root.It’s function is cooling heat and blood, detoxicating and mois-tening.There are many chemical composition in the Radix Scrophular-iae such as harpagide, acubin, catapol, harpagoside,6’-O-acetyl harpagoside, angroside C,8-O-(2-hydroxycinnamoyl)-harpagide, geniposide and so on.In this paper, we studied in36different origins preparedslices of Radix Scrophulariae by HPLC characteristic spectrum and constast the difference of single wavelength and multiwavel-ength to analysis the results.At last, we found the most appro-priate chromatographic separation conditions, chromatographic column:Themorn C18(5μm,4.6mm×250mm),column temperature:30℃,velocity:1ml/min, acetonitrile and0.1%formic acid gradientelution:0~15min,3%~10%acetonitrile;15~30min acetonitrile,10%~25%acetonitrile,30~45min,25%~35%acetonitrile;45~50min,35%~3%acetonitrile, detected multiwavelength:0~5min230nm;5~13min210nm;13~23min320nm;23~50min278nm,sample weight:20μlThis condition could makethe characteristiccomponents of Radix Scrophulariae such as harpagide, acubin, catapol, harpagoside and cinnamic acid separate better andshown in the chromatogram.According to comparing the different HPLC CharacteristicSpectrum, we take the peak of hapagoside as the standard oneand calculation the relative retention time, relative peakareaand similarity and detection rate in every sample to evaluatingthe quality. Considering the prepared slices of Radix Scrophulariae claim of<Chinese Pharmacopoeia>, we select10best sampleto established the superior grade standard HPLC CharacteristicSpectrum to distinguish differentkinds of sample from the rela-tive retention time, relative peak area and similarity and dete-ction rate. According the result, the number of similarity hig-her than0.70is35andthe highest is0.990; the number higherthan0.93is16, between0.85-0.93is12and8smaller than0.85. So we decided the similarity of prepared slices of RadixScrophulariae must higher than0.70. The excellent lever piecesshould not be less than0.93, the A pieces should not be lessthan0.85and the EC good should not be less than0.70.Studying and comparing with the two different detectionconditions, we based on the single wavelength and improved the meth-od,we builded the multiwavelength method which could reflectthe distinct in different quality of prepared slices of RadixScrophulariae comprehensive.In this report, we established thesuperior grade standard HPLC Characteristic Spectrum that prov-ide a reference to control the quality of it.
