Polygonum multiflorum,commonly used traditional Chinese medicine is dry root which stems from plant tuber of knotweed.Polygonum multiflorum thunb with flat qualities and a little bitter,sweet flavor is good at disposing hotness,poison,subside swelling as well as bump.Hence,it is usually used as clinical turgescence and laxative.When it has been prepared,Polygonum multiflorum becomes warmer and sweeter.It could make hair dark and people strong.Furthermore,prepared Polygonum multiflorum could increase the ability of immunity.modern chemistry and pharmacodynamics study confirms that Polygonum multiflomm cosistent of two major components:one is stilbene glucoside which has effects of anti-aging,lowering cholesterol,improving immune function,prevention and treatment of atherosclerosis and the role of the liver,etc.The other component is anthraquinones which plays an important role in anti-bacterial,anti-tumor,myocardial injury,bleeding and other biological activity.In view of the complexity of Chinese herbal ingredients and the characteristics of clinical usage(pieces often used directly or combined preparation process is relatively simple preparation of medicines) multiple indicator determination of composition evaluation of Chinese herbal medicines has become the development trend of quality standards.Compared to single indicator of the determination of composition in comparison,multi-index components is a more comprehensive refection of the intrinsic quality of medicines.Also it could help to improve the specific quality standards.In order to improve the quality standards of objectivity and specificity in Polygonum multiflorum medicine,a novel method was proposed in this paper.It utilized stilbene glycosides and anthraquinones which are the major components of Polygonum multiflorum as an index.The determination methods,different regions and markets determination and variance analysis,different growth period and harvesting,processing technique of Polygonum multiflorum were reappraised.Some results achieved as follows:1 A HPLC method was established for the determination of 2,3,5,4'-tetrahydroxystilbene-2-O-β-D-glucopyranoside and anthraquinon simultaneously in Radix Polygonum multiflori.The attractive feature of this method was reduction of sample steps and operation of the system error.Compared to the separate components of the indicators measure approach it was simple,accurate and practical.2 In our experiment,stilbene glycosides and anthraquinones(emodin,physcion, emodin glucoside,physcion glucoside)contents in wild Polygonum multiflorum from 17 origins of 6 provinces and municipalities including Guangdong,Hunan,Guangxi, Guizhou,Chongqing,Hubei and so on.The results showed that for samples of Polygonum multiflorum Thunb in different origins,there was no significant correlation in the contents of stilbene glycosides and anthraquinones.Considering their different pharmacological activity,cluster analysis were carded out for 17 different production batches of stilbene glucoside of Polygonum multiflorum and anthraquinones using SPSS15.0(SPSS Inc.).According to the results,we make a conclusion that the cultivation place of Polygonum multiflorum should be dependent of medical use.Chinese herbal medicine for the benefit should be the preferred type of Laibin,Guangxi and Longsheng,Guangxi for its high-quality of stilbene glucoside. If used as a type of diarrhea medicine,the ones planted in higher anthraquinones origin like Guiding Guizhou,Dujun Guizhou are optional.3 Active ingredients of Polygonum multiflorum of ten different approved products was measured.Relevant quality standards were set according to the active ingredients in Polygonum multiflorum from different regions in conjunction with water,ash,acid insoluble ash and so on in samples.4 In order to investigate the dynamic changes between chemical ingredients and Polygonum multiflorum Thunb seasonal growth,the 1~3 years Polygonum multiflorum collected from specific place(Yuelai Shizhu production base,Chongqing Institute of Chinese Medicine) in July-december were studied by our method.The results showed that for 1~3- year-old stem of Polygonum multiflorum Thunb,the contents of active ingredients achieve the maximum in the mid November.Therefore, we infer that the best harvest time for Polygonum multiflorum is in mid November each year.While for 1~4 -year-old Polygonum multiflorum Thunb,the active ingredients of 3-year-old has reached the highest value.5 Polygonum multiflorum quality changes in processing and storage were studied systematically.The results showed that the thickness of different sections had have little effect to contents of active ingredients,whereas,Polygonum multiflorum storaged in bags could maintain the highest content after a period of time.Also different drying temperature especially at 60℃and drying method had big impact on the contents of active ingredients.Based on the findings 2-4mm of the thin,60℃drying or drying indoor and storage in bags would be a good storage method.
