ObjectiveTo explore the etiological factor of senility and observe the effects of reinforcing the kidney, promoting blood circulation and removing phlegm therapy on APN, 8-OHdG, T-AOC in blood serum and apoptosis of hippocampus and kidney for analyzing its mechanism of action for post- poning senility. And to observing the effect of postponing senility in different time through setting up force-medication administration group and post-medication administration group.Methods1. Grouping: Sixty 3-month-old around male SD rats were randomly divided into six groups: normal group, senility model group, VitE treated group, experiment (prescription of reinforcing the kidney, promoting blood circulation and removing phlegm treated) group, force-medication administration group and post-medication administration group.2. Molding: Execpt the normal group, the others induced by injection of back with D-galactose to make senility model.3. Giving medicine: Normal group and senility model group with 0.9% NS everyday; experiment group with prescription everyday; VitE treated group with VitE everyday; force-medication administration group with prescription everyday the first three weeks and with 0.9% NS everyday the last three weeks; post-medication administration group with 0.9% NS everyday the first three weeks and with prescription everyday the last three weeks.The dosage administed 1ml/100g.4. Detection: The contents of APN, 8-OHdG, T-AOC in blood serum and apoptosis of hippocampus and kidney.Results1. Apoptosis of hippocampus: Normal group and experiment group were significantly lower than senility model group(P<0.01). VitE treated group and force-medication administration group were lower than senility model group (P<0.05). VitE treated group and post-mdication administration group were higher than experiment group (P<0.05). Apoptosis of kidney: Normal group and experiment group were lower than senility model group(P<0.05).2. The contents of APN in blood serum: Normal group and experiment group were significantly higher than senility model group (P<0.01). VitE treated group was higher than senility model group (P<0.05). Post-mdication administration group was lower than experiment group (P<0.05).3. The contents of 8-OHdG in blood serum: Normal group, experiment group, VitE treated group and force-medication administration group were significantly lower than senility model group(P<0.01).Post-medication administra-tion group was lower than senility model group(P<0.05). Experiment group and force-medication administration group were lower than post-medication administration group (P<0.05).4. The contents of T-AOC in blood serum: Normal group was significantly higher than senility model group(P<0.01). Experiment group, VitE treated group and force-medication administration group were higher than senility model group(P<0.05). Experiment group was higher than VitE treated group(P<0.05).Conclusion1. Kidney weak, blood stasis and phlegm is the basic etiological factor for senility, and reinforcing the kidney, promoting blood circulation and removing phlegm therapy is operative on postponing senility.2. The Prescription of reinforcing the kidney, promoting blood circulation and removing phlegm possibly possesses a certain improvement of the APN and T-AOC content in blood serum, decreased the 8-OHdG content in blood serum, suppression the apoptosis of hippocampus and kidney for senility model rats, consequently postponing senility.3. The effects on improvement of the APN content in blood serum, decreased the 8-OHdG content in blood serum and suppression the apoptosis of hippocampus in experiment group are more conspicuous than that of post-mdication administration group. The content of 8-OHdG in blood serum is lower in force-medication administration group than that of post-medication administration group obviously.And to postpone senility by decreasing or recoverying oxidative DNA damage.
